摘  要:目的:探讨色素上皮衍生因子(PEDF)联合5-氟尿嘧啶(5-FU)对结肠癌CT26细胞的抑制作用以及对荷瘤小鼠模型的治疗效果。方法:将体外培养的CT26细胞分为对照组、5-FU组和PEDF+5-FU组3组,分别加入PBS、5-FU和PEDF+5-FU处理CT26细胞。通过细胞划痕实验和Transwell小室检测CT26细胞迁移能力。建立CT26荷瘤小鼠模型,将24只荷瘤模型小鼠随机分为对照组、5-FU组和PEDF+5-FU组,每组8只。用相应药物给药干预,共给药3次,20 d后处死小鼠测量肿瘤体积。通过CD31免疫荧光染色检测肿瘤组织内的微血管密度,TUNEL染色观察肿瘤细胞凋亡情况。结果:与对照组比较,5-FU组和PEDF+5-FU组CT26细胞迁移率显著下降,CD31表达明显下调,肿瘤细胞的凋亡率明显提高(均P<0.05);与5-FU组相比,PEDF+5-FU组CT26细胞迁移率显著下降,CD31表达明显下调,肿瘤细胞凋亡率明显提高(P<0.05)。与对照组相比,5-FU组和PEDF+5-FU组肿瘤体积显著减小(均P<0.05),PEDF+5-FU组肿瘤体积小于5-FU组(P<0.05)。结论:PEDF和5-FU联合用药可以对CT26结肠癌细胞起到协同治疗的目的,且治疗效果优于5-FU单独治疗效果。Objective:To investigate the inhibitory effect of pigment epithelium-derived factor(PEDF)combined with 5-fluorouracil(5-FU)on colon cancer CT26 cells and the therapeutic effect on tumor-bearing mice.Methods:CT26 cells cultured in vitro were divided into control group,5-FU group and PEDF+5-FU group.Thethree groups were treated with PBS,5-FU and PEDF+5-FU,respectively.Cell scratch test and Transwell chamber test were used to detect the difference of migration ability of CT26 cells in each group.The model of CT26 tumorbearing mice was established.24 tumor-bearing mice were randomly divided into three groups:control group,5-FU group and PEDF+5-FU(n=8 per group).The mice were treated with drugs for 3 times.20 days later,the mice were euthanatized,and the volume of the tumor was measured.The expression of neovascularization CD31 and apoptotic cells in tumor tissue were evaluated by immunohistochemistry and immunofluorescence,respectively.Results:Compared with the control group,the migration ability of CT26 cells was significantly decreased,the expression of CD31 down-regulated and the apoptosis rate of tumor cells were significantly increased in 5-FU group and PEDF+5-FU group(P<0.05).Compared with the 5-FU group,the migration ability of CT26 cells was significantly decreased,the expression of CD31 down-regulated and the apoptosis rate of tumor cells were significantly increased in PEDF+5-FU group(P<0.05).Compared with the control group,tumor volume was markedly reduced in 5-FU group and PEDF+5-FU group(P<0.05),the tumor volume in PEDF+5-FU group was smaller than that in 5-FU group(P<0.05).Conclusion:The combination of PEDF and 5-FU can synergistically treat CT26 colon cancer cells.The synergistic effect is better than curative effect of 5-FU alone.
