当归补血汤中毛蕊异黄酮葡萄糖苷及阿魏酸的含量测定

摘　　要：目的建立同时测定当归补血汤中两种指标性成分-毛蕊异黄酮葡萄糖苷、阿魏酸含量的HPLC方法。方法使用Agilent 5TC-C18色谱柱,4.6 mm×250 mm,5μm;流动相A:乙腈,B:0.02 mol/L磷酸二氢钾溶液(用磷酸调节pH值至3.0),梯度洗脱(0~6 min,98%B,6~15 min,98%~80%B,15~30 min,80%B,30~50 min,80%~60%B,50~60 min,60%~35%B,60~75 min,35%B,75~80 min,35%~98%B),流速:1.0 ml/min;波长:280 nm;柱温:30℃;检测毛蕊异黄酮葡萄糖苷、阿魏酸含量。结果阴性对照无干扰,毛蕊异黄酮葡萄糖苷、阿魏酸的线性范围分别为5.83~58.29μg/ml(r=0.9999),3.03~30.26μg/ml(r=0.9995)其平均加样回收率分别为102.25%和98.63%。结论所建含量测定方法简便、准确和可靠,可作为当归补血汤质量控制的标准。Objective To establish a high-performance liquid chromatography(HPLC)method for the simultaneous determination of two index components,which were calycosin glucoside and ferulic acid,in Danggui Buxue Decoction.Methods Agilent 5TC-C18 column(4.6 mm×250 mm,5μm)was used.The mobile phase A was acetonitrile and B was 0.02 mol/L potassium dihydrogen phosphate solution(pH value was adjusted to 3.0 with phosphoric acid).The gradient elution time was 0-6 min with 98%B,6-15 min with 98%-80%B,15-30 min with 80%B,30-50 min with 80%-60%B,50-60 min with 60%-35%B,60-75 min with 35%B and 75-80 min with 35%-98%B.The flow rate was 1.0 mL/min,the wavelength was 280 nm,and the column temperature was 30℃.The contents of calycosin glucoside and ferulic acid were detected.Results The linear ranges of calycosin glucoside and ferulic acid were 5.83-58.29μg/mL(r=0.9999)and 3.03-30.26μg/ml(r=0.9995),and the average sample recovery rates were 102.25%and 98.63%,respectively.Conclusion The established method is simple,accurate and reliable,which can be used as the quality control standard of Danggui Buxue Decoction.
