摘　　要：目的分析Fcγ受体3A(FCGR3A)在脑胶质瘤中的表达特点及其临床意义。方法选取中国脑胶质瘤基因组图谱数据库中1018例脑胶质瘤患者的转录组数据及临床资料,同时选用癌症基因组图谱数据库中668例脑胶质瘤患者的数据进行验证,分析FCGR3A在脑胶质瘤中的表达特征。采用Kaplan-Meier生存曲线和Cox回归分析FCGR3A相对表达量对脑胶质瘤患者生存时间的影响。通过基因本体(GO)功能富集分析FCGR3A及其相关基因的生物学功能;对患者的转录组数据进行免疫浸润评分,探究FCGR3A相对表达量与肿瘤免疫浸润程度的关系。结果FCGR3A相对表达量随脑胶质瘤恶性程度上升而增加,在高恶性程度的胶质母细胞瘤、异柠檬酸脱氢酶野生型胶质瘤和间质型胶质瘤均高表达。FCGR3A高表达患者总生存期显著低于低表达组,单因素及多因素Cox回归显示FCGR3A高表达是胶质瘤患者总生存期的独立危险因素。GO分析显示FCGR3A参与免疫细胞激活等多种免疫过程,免疫浸润分析显示FCGR3A相对表达量与胶质瘤内免疫细胞浸润程度正相关。结论FCGR3A表达与脑胶质瘤恶性程度正相关,可作为胶质瘤患者预后的预测指标及潜在的治疗靶点。Objective To analyze the expression characteristics of FCGR3A in glioma and its clinical significance.Methods The transcriptome data and clinical data of 1018 glioma patients from the Chinese Glioma Genome Atlas database were included,and the data of 668 glioma patients was selected for verification from the Cancer Genome Atlas database.The expression characteristics of FCGR3A were analyzed in different groups.Kaplan-Meier survival curve and Cox regression were used to analyze the effect of FCGR3A expression on the overall survival of patients with glioma.The biological functions of FCGR3A and its related genes were evaluated by gene ontology(GO)functional enrichment.The immune infiltration score was performed on the transcriptome data of the patients to explore the relationship between the expression level of FCGR3A and the degree of tumor immune infiltration.Results The expression of FCGR3A increased with the malignant progression of glioma.FCGR3A showed high expression in the glioblastoma,the IDH wild type and the mesenchymal type.The overall survival of patients with high FCGR3A expression was significantly lower than that of the low expression group.Univariate and multivariate Cox regression analysis found that the high expression of FCGR3A was an independent risk factor for the overall survival of patients with glioma.FCGR3A-related genes were mainly enriched in the process of multiple immune cell activation.The increase of FCGR3A expression was also positively correlated with the increase of immune cell infiltration.Conclusion The expression level of FCGR3A is positively correlated with the malignant degree of glioma,and can be used as a prognostic indicator and potential therapeutic target for gliomas.
