摘　　要：目的:探讨miR-614过表达对肺腺癌A549细胞增殖、侵袭及裸鼠移植瘤生长的影响。方法:将培养至对数生长期的人A549细胞分为对照组、miR-NC组(脂质体转染miR-NC mimic)和miR-614组(脂质体转染miR-614 mimic)。RT-qPCR检测每组A549细胞中miR-614的表达,CCK-8法检测每组A549细胞增殖活性;Transwell法检测检测每组A549细胞侵袭细胞数,Western Blot法检测每组A549细胞血管内皮生长因子(VEGF)、基质金属蛋白酶(MMP)-2、金属蛋白酶组织抑制因子-2(TIMP-2)和Ki-67蛋白的相对表达量。皮下种植0.2 mL各组A549细胞悬液(5×10^(6)/mL),构建裸鼠移植瘤的模型,每周测量1次移植瘤组织的体积,造模5周后进行移植瘤组织称重。结果:miR-614组A549细胞增殖活性和侵袭细胞数低于miR-NC组(P<0.05),细胞VEGF、MMP-2和Ki-67蛋白表达低于miR-NC组(P<0.05),TIMP-2蛋白表达高于miR-NC组(P<0.05);miR-614组裸鼠移植瘤质量低于miR-NC组,且造模3周、4周和5周后,移植瘤体积低于miR-NC组(P<0.05)。结论:miR-614过表达可抑制A549细胞的增殖和侵袭,进而抑制肿瘤生长。Objective:To explore the effects of miR-614 overexpression on proliferation and invasion of lung adenocarcinoma A549 cells and growth of transplanted tumor in nude mice.Methods:Human A549 cells were divided into control group,miR-NC group(liposome was transfected with miR-NC mimic)and miR-614 group(liposome was transfected with miR-614 mimic).The expression of miR-614 in each group was detected by RT-PCR.The proliferation activity of cells was detected by CCK-8.The cells invasion was detected by Transwell.The expression of vascular endothelial growth factor(VEGF),matrix metalloproteinase(MMP)-2,tissue inhibitor of metalloproteinase-2(TIMP-2)and Ki-67 protein were detected by Western blot.The subcutaneous implantation of 0.2 mL A549 cells suspension(5×10^(6)/mL)in each group was conducted to establish the transplanted tumor model of nude mice.The tumors volume was measured once a week,which were weighed at 5 weeks after molding.Results:The proliferation activity and number of invasive A549 cells in miR-614 group were lower than those in miR-NC group(P<0.05),the expression of VEGF,MMP-2 and Ki-67 protein were lower than those in miR-NC group(P<0.05),while expression of TIMP-2 protein was higher than that in miR-NC group(P<0.05).The mass of transplanted tumors in miR-614 group was lower than that in miR-NC group,and volume of transplanted tumor was smaller than that in miR-NC group at 3,4 and 5 weeks after molding(P<0.05).Conclusion:Overexpression of miR-614 will inhibit the proliferation and invasion of A549 cells,and inhibit tumor growth.
