摘 要:目的:在基因表达谱的水平分析头颈部肿瘤鼻咽癌的差异表达基因(DEGs),鉴定候选基因及信号通路。方法:依据Home-GEO-NCBI数据库中的高通量基因表达数据,对GEO数据集进行统计。以GEO2R、David、Amigo、Kegg-Pathway、Reactome、Panther等作为基础,整合四个芯片数据集数据(GSE12452,GSE13597,GSE53819和GSE64634),统计四组数据及DEGs,并进行GO基因功能富集分析、KEGG信号通路富集分析和蛋白相互作用网络分析。结果:本研究发现114个有统计学意义的DEGs(38个上调基因和76个下调基因)。通过基因功能富集分析及信号通路富集分析,发现36个中心节点基因(FN1、CD19、C3、CXCL10、CLU、PROM1、CD22、MMP3、CR2、SCGB1A1、MEF2C、PAX5、PIGR、GAD1、PLAU、MAD2L1、MSLN、VCAN、DST、HOXA10、LTF、MS4A1、MUC16、PTGDS、ANG、SPAG6、TCL1A、TEKT2、TSPAN8、ATP12A、BANK1、CR1、DNAI1、KLF2、KRT7、NRXN1)。上述基因富集于癌症中的转录失调、细胞粘附分子(CAMs)、PI3K-Akt信号通路等过程。结论:中心节点基因与鼻咽癌发生、发展密切相关,可以为鼻咽癌的早期诊断及靶向治疗提供潜在线索。Objective:To analyze the differentially expressed genes(DEGs)of head and neck tumors and nasopharyngeal carcinoma(NPC)at the level of gene expression profile,and to identify candidate genes and signaling pathways.Methods:According to the high-throughput gene expression data in Home-GEO-NCBI database,the GEO data set was statistically analyzed.Based on Geo2R,David,Amigo,KEGG-Pathway,Reactome,Panther,etc.,the data of four chip datasets(GSE12452,GSE13597,GSE53819 and GSE64634)were integrated,and the four sets of data and DEGS were analyzed.The GO gene function enrichment analysis,KEGG signaling pathway enrichment analysis and protein interaction network analysis were performed.Results:114 DEGs(38 up-regulated genes and 76 down-regulated genes)were found with statistical significance.Through gene function enrichment analysis and signal pathway enrichment analysis,36 central node genes(FN1,CD19,C3,CXCL10,CLU,PROM1,CD22,MMP3,CR2,SCGB1A1,MEF2C,PAX5,PIGR,GAD1,PLAU,MAD2L1,MSLN,VCAN,DST,HOXA10,LTF)were found,(MS4A1,MUC16,PTGDS,ANG,SPAG6,TCL1A,TEKT2,TSPAN8,ATP12A,BANK1,CR1,DNAI1,KLF2,KRT7,NRXN1).These genes are enriched in transcriptional disorders,cell adhesion molecules(CAMs),PI3K-Akt signaling pathway and other processes in cancer.Conclusion:The central node gene is closely related to the occurrence and development of nasopharyngeal carcinoma,which may provide potential clues for the early diagnosis and targeted therapy of nas opharyngeal carcinoma.
