摘 要:目的探究苦豆碱对胃癌细胞生长的抑制作用。方法分别用0 mmon·L^(-1)、1 mmon·L^(-1)、2 mmon·L^(-1)和4 mmon·L^(-1)的苦豆碱处理SGC7901和BGC823细胞0、12、24、36和48 h,通过MTT实验检测苦豆碱对细胞增殖能力的影响;通过流式细胞术及Annexin V-PI染色检测苦豆碱对SGC7901细胞周期和凋亡能力的影响;通过western blot检测苦豆碱对SGC7901细胞中CDK4、CDK6和bcl-2表达水平的影响。结果1 mmmon·L^(-1)、2 mmon·L^(-1)和4 mmon·L^(-1)的苦豆碱作用于SGC7901细胞24 h时对细胞增殖的抑制率分别为(26.41±2.96)%、(41.51±4.45)%和(62.88±4.59)%,1 mmol·L^(-1)、2 mmol·L^(-1)和4mmol·L^(-1)苦豆碱作用于BGC823细胞24 h时对细胞增殖的抑制率分别为(28.35±1.86)%、(40.11±2.65)%和(61.77±3.84)%,与0 mmon·L^(-1)相比差异均具有统计学意义;苦豆碱还可抑制细胞周期G1/S期的进程,促进细胞凋亡;Western blot结果显示,苦豆碱可抑制CDK4、CDK6和bcl-2的表达。结论苦豆碱可抑制胃癌细胞的增殖促进其凋亡。Objective To investigate the inhibitory effect of aloperine on the growth of gastric cancer cells. Methods SGC7901 and BGC823 cells were treated with 0 mM, 1 mM, 2 mM and 4 mM aloperine for 0, 12, 24, 36 and 48 h respectively. MTT assay was used to detect the effects of aloperine on cell proliferation, flow cytometry and Annexin V-PI staining were used to detect the effects of aloperine on cell cycle and apoptotic capacity of SGC7901 cells. Western blot was used to detect the effects of aloperine on the expression of CDK4, CDK6 and bcl-2 in SGC7901 cells. Results The inhibition rates of 1 mM, 2 mM and 4 mM aloperine on cell proliferation of SGC7901 cells at 24 h were(26.41±2.96)%,(41.51±4.45)% and(62.88±4.59)% respectively. The inhibition rates of 1,2 and 4 mmon·L^(-1) aloperine on cell proliferation of BGC823 cells at 24 h were(28.35±1.86)%,(40.11±2.65)% and(61.77±3.84)% respectively, compared with 0 mM, the difference was statistically significant. Aloperine can also inhibit the G1/S phase of cell cycle and promote cell apoptosis. The results of western blot showed that aloperine could inhibit the expression of CDK4, CDK6 and bcl-2. Conclusion Aloperine can inhibit the proliferation of gastric cancer cells and promote their apoptosis.
