摘  要:采用大孔树脂分离纯化纵条纹炭角菌中环肽Xylastriamide A。以HPLC法测定纵条纹炭角菌中环肽Xylastriamide A含量,通过比较4种不同型号的大孔树脂对该环肽吸附-解吸效果及静态动力学研究,确定D101大孔树脂为最佳的纯化材料。通过单因素试验确定D101大孔树脂对纵条纹炭角菌中环肽Xylastriamide A的吸附-解吸效果最佳工艺条件为粗提物以质量浓度3 mg/mL、吸附流速2 BV/h上样7 BV,以40%乙醇3 BV洗去杂质后,用100%乙醇3 BV以流速为2 BV/h洗脱。在此工艺条件下,环肽Xylastriamide A纯度达到39.63%,表明D101大孔树脂对纵条纹炭角菌中环肽Xylastriamide A具有较好的纯化效果。该工艺稳定可靠,为环肽Xylastriamide A的纯化提供方法,也为其他类型的环肽成分的分离纯化提供思路。The cyclopeptide Xylastriamide A was isolated and purified with macroporous resin.The HPLC method was used to determine the content of cyclopeptide Xylastriamide A in Xylaria striata.By comparing the adsorption and desorption effects of four different types of macroporous resin on the cyclopeptide and the static kinetics study,D101 macroporous resin was determined as the best purified material.Single factor test was conducted to determine the optimal adsorption/desorption effect of D101 macroporous resin on the cyclopeptide Xylastriamide A in Xylaria striata.The optimum condition was as follows:the crude extract was sampled with A mass concentration of 3 mg/mL and an adsorption flow rate of 2 BV/h;the impurities were washed with 40%ethanol 3 BV,and eluted with 100%ethanol 3 BV with A flow rate of 2 BV/h.Under this process condition,the purity of cyclopeptide Xylastriamide A reached 39.63%,indicating that D101 macroporous resin had a good purification effect on the cyclopeptide Xylastriamide A in Xylaria striata.This process is stable and reliable,which provides A method for the purification of cyclopeptide Xylastriamide A,and also provides an idea for the separation and purification of other cyclopeptide components.
