摘  要:蛹虫草多糖具有抗氧化、抗肿瘤等多种药理活性,在食药用方面有很好的发展前景.植物多糖因含有色素和酚类化合物,导致颜色较深,不利于产品的开发.因此,对蛹虫草粗多糖的脱色纯化工艺的研究势在必行.文章以蛹虫草多糖的保留率、脱色率以及蛋白清除率为指标,通过单因素方差试验初步确定提取过程的主要影响因素,再设计中心组合试验,得到最佳工艺参数:上样溶液体积为175 mL,质量浓度为1.25 mg/mL,控制pH为5.5,常温下以1.5 BV/h的流速上样,收集多糖脱色溶液.在上述条件下,重复进行3次验证试验,得到溶液的多糖保留率为:85.87%±0.83%,脱色率为:70.57%±1.26%,蛋白清除率为:96.93%±0.75%.Cordyceps militaris polysaccharide has various pharmacological activities such as anti-oxidation and anti-tumor,so it has good development prospects in food and medicine.Plant-derived polysaccharides contain pigments and phenolic compounds,resulting in darker colors,which is not conducive to the development of related products.Therefore,it is imperative to study the decoloration and purification process of Cordyceps militaris crude polysaccharide.Based on the retention rate,decolorization rate and protein clearance rate of Cordyceps militaris polysaccharide,the main influencing factors of the extraction process are preliminarily determined through a single-factor variance test,and then a central combination test is designed to obtain the best process parameters.The result was as follows:the volume of the sample solution is 175 mL,the mass concentration is 1.25 mg/mL,the pH is controlled to 5.5,the sample is loaded at a flow rate of 1.5 BV/h at room temperature,and the polysaccharide decolorizing solution is collected.Under the above conditions,the verification test was repeated 3 times,and the polysaccharide retention rate of the solution was 85.87%±0.83%,the decolorization rate was 70.57%±1.26%,and the protein clearance rate was 96.93%±0.75%.
