摘  要:为了明确马铃薯杂交组合‘MH-09’בMIN-21’F_(1)代分离群体中选育出的8个优良杂种株系间的分子细胞遗传学差异,为后续新品种育成及鉴定提供依据,利用亲本材料作对照,对其花粉育性、PMCMⅠ染色体配对行为及SRAP(Sequence related amplified polymorphism,序列相关扩增多态性)指纹特征等进行了比较研究.结果表明:各杂种株系间的花粉可育率存在一定差异,变幅为48.1%~77.3%,株系ZX-1540的花粉育性低于50%,其余株系均在55%以上;8个杂种株系ZX-20、ZX-460、ZX-780、ZX-900、ZX-1120、ZX-1540、ZX-1960和ZX-2200的PMCMⅠ染色体配对构型分别是3.82Ⅰ+13.05Ⅱ+2.31Ⅲ+2.79Ⅳ,6.52Ⅰ+13.04Ⅱ+2.24Ⅲ+2.17Ⅳ,4.37Ⅰ+9.97Ⅱ+3.19Ⅲ+3.53Ⅳ,4.05Ⅰ+15.54Ⅱ+2.12Ⅲ+1.63Ⅳ,2.71Ⅰ+13.85Ⅱ+2.35Ⅲ+2.64Ⅳ,8.44Ⅰ+7.08Ⅱ+5.92Ⅲ+1.91Ⅳ,5.20Ⅰ+8.61Ⅱ+6.76Ⅲ+1.33Ⅳ和1.13Ⅰ+11.69Ⅱ+1.04Ⅲ+5.09Ⅳ;利用筛选出的10对SRAP适宜引物,对10个材料进行PCR扩增得到122个多态性位点条带,多态性比率为65.92%;用特异性引物M5BE7扩增建立了能鉴定8个优良杂种株系和其亲本的SRAP指纹图.各株系及其亲本的遗传距离(GD)范围在0.290~0.640之间.以GD值0.40作基准,将10个材料划分成5类:株系ZX-20、ZX-1960和母本‘MH-09’为一类;株系ZX-780、ZX-1120、ZX-2200和父本‘MIN-21’为一类;株系ZX-460、ZX-900和ZX-1540各为一类.In order to identify the molecular cytogenetic differences between eight excellent hybrid strains selected from the F_(1) potato hybrids of‘MH-09’בMIN-21’,and provide a basis for the breeding and identification of subsequent new varieties,in this research,using the parent materials as control,we compared and analyzed the pollen fertility,behavior of PMCM I chromosome pairing and SRAP(Sequence related amplified polymorphism)fingerprint characteristics of the excellent hybrid strains.The results showed that there were some differences in pollen fertility rates among these hybrid strains,the variation ranged from 48.1%to 77.3%.The pollen fertility of strain ZX-1540 was less than 50%,and the rest of the strains were above 55%.The chromosome paired configurations(PMCMI)of the eight hybrid strains ZX-20,ZX-460,ZX-780,ZX-900,ZX-1120,ZX-1540,ZX-1960 and ZX-2200 are 3.82Ⅰ+13.05Ⅱ+2.31Ⅲ+2.79Ⅳ,6.52Ⅰ+13.04Ⅱ+2.24Ⅲ+2.17Ⅳ,4.37Ⅰ+9.97Ⅱ+3.19Ⅲ+3.53Ⅳ,4.05Ⅰ+15.54Ⅱ+2.12Ⅲ+1.63Ⅳ,2.71Ⅰ+13.85Ⅱ+2.35Ⅲ+2.64Ⅳ,8.44Ⅰ+7.08Ⅱ+5.92Ⅲ+1.91Ⅳ,5.20Ⅰ+8.61Ⅱ+6.76Ⅲ+1.33Ⅳ,and 1.13Ⅰ+11.69Ⅱ+1.04Ⅲ+5.09Ⅳ,respectively.Using 10 screened SRAP suitable primers to amplify the 10 materials,a total of 122 polymorphic bit bands were obtained,with a polymorphism ratio of 65.92%.Based on the specific primer M5BE7,a SRAP fingerprint map was constructed to identify 8 excellent hybrid strains and their parents.The genetic distance(GD)of each strain and its parent ranged from 0.290 to 0.640.Using the GD value of 0.40 as a standard,the 10 materials were divided into five categories:strain ZX-20,ZX-1960 and mother‘MH-09’as a category;strain ZX-780,ZX-1120,ZX-2200 and father‘MIN-21’as a category;strain ZX-900 and ZX-1540 as one category,respectively.
