摘　　要：目的:研究人载脂蛋白M(apolipoprotein M,Apo M)对肾透明细胞癌细胞株(ACHN、769-P和786-O)增殖、迁移和侵袭能力的影响。方法:采用装载人Apo M基因的慢病毒和阴性对照组慢病毒分别感染肾癌细胞株,建立Apo M过表达组(Apo M-OE组)和阴性对照组(NC组)细胞模型。应用CCK-8增殖实验和克隆形成实验观察细胞增殖能力,采用细胞划痕实验检测细胞迁移能力,采用Transwell实验检测细胞侵袭能力。结果:Apo M过表达后,ACHN和769-P过表达组细胞在72h增殖能力显著降低(分别P<0.01,P<0.05);786-O细胞在48 h表现出明显差异(P<0.001)。克隆形成实验表明Apo M过表达后,3株细胞克隆形成能力明显减弱(分别P<0.001,P<0.01,P<0.01)。细胞划痕实验证实ACHN和769-P过表达组细胞在12 h时出现明显差异(分别P<0.05,P<0.001);786-O细胞在24h时迁移能力明显受抑(P<0.01)。细胞侵袭实验发现3株细胞过表达A po M后侵袭能力下降(分别P<0.001,P<0.001,P<0.01)。结论:Apo M过表达后,肾癌细胞增殖、迁移和侵袭能力显著减弱。Objective:To investigate the effect of apolipoprotein M(Apo M)on the proliferation,migration,and invasion of renal clear cell carcinoma cell lines(ACHN,769-P,and 786-O).Methods:To establish Apo M overexpression(Apo M-OE)cell models,lentivirus carrying human Apo M gene and negative control(NC)lentivirus were used to infect renal clear cell carcinoma cell lines respectively.CCK-8 assay and colony formation assay were employed to evaluate the proliferation capacity of renal cancer cells.Wound healing assay was used to detect the migration capacity of tumor cells and transwell assay was applied to examine the invasion ability.Results:The proliferation capacity of Apo M-OE groups was significantly reduced at 72 h in ACHN and 769-P cells(P<0.01,P<0.05,respectively).The difference in 786-O cell arises at 48 h(P<0.001).The colony formation assay indicated that the capacity of colony formation was significantly reduced in 3 renal carcinoma cell lines(P<0.001,P<0.01,P<0.01,respectively).Wound healing assay showed that the migration of Apo M-OE groups in ACHN and769-P was restrained at 12 h(P<0.05,P<0.001,respectively);the migration of 786-O Apo M-OE group was suppressed at 24 h(P<0.01).Transwell assay found that the invasion of renal carcinoma cells was inhibited due to Apo M overexpression(P<0.001,P<0.001,P<0.01,respectively).Conclusion:Apo M significantly inhibites the proliferation,migration,and invasion capacity of renal carcinoma cells.
