摘 要:目的应用生物信息学方法初步探讨PARPBP基因在结肠癌中的表达及预后价值。方法通过TCGA数据库下载结肠腺癌(COAD)数据集,利用差异分析和HPA数据库考察COAD中PARPBP的mRNA和蛋白表达水平。借助WebGestalt在线工具对PARPBP表达相关基因进行KEGG富集分析。采用Log-rank法和卡方检验考察PARPBP与COAD患者预后及临床病理特征的相关性。在GEO数据库中分析PARPBP与COAD化疗耐药之间的关系。结果COAD组织中PARPBP的mRNA和蛋白表达水平均上调。功能富集分析发现,PARPBP表达相关基因主要富集于DNA损伤修复和DNA复制相关通路,通过调控染色体不稳定性参与结肠癌的发生。此外,PARPBP基因低表达与COAD患者较高的病理N分期、肿瘤分期和不良预后显著相关。PARPBP基因在药物抵抗的结肠癌细胞株中表达下调,说明其与COAD细胞的药物敏感性相关。结论PARPBP在结肠癌组织中高表达,其高表达是结肠癌患者预后的保护性因素。Objective To preliminarily explore the expression and prognostic value of PARPBP in colon cancer by bioinformatics analysis.Methods The data sets of colon adenocarcinoma(COAD)were downloaded from the TCGA database.Then the mRNA and protein expression of PARPBP in COAD were investigated with the differential analysis and HPA database.The KEGG enrichment analysis of PARPBP-related genes was performed by WebGestalt online tool.Log-rank and chi-square method were used to investigate the correlation of PARPBP expression with prognosis and clinicopathological characteristics of COAD patients.Finally,the expression of PARPBP in chemoresistant COAD cells was detected in the GEO database.Results The mRNA and protein expression levels of PARPBP were upregulated in COAD tissues.Functional enrichment analysis showed that PARPBP-related genes were mainly enriched in pathways comprising DNA damage repair and DNA replication,and participated in the occurrence of colon cancer by regulating chromosomal instability.In addition,the low expression of PARPBP was significantly related to the higher pathological N stage,tumor stage and poor prognosis of COAD patients.Together with the finding of PARPBP down-regulation in drug-resistant colon cancer cell lines,our results indicated that PARPBP was related to the drug sensitivity of COAD cells.Conclusion PARPBP is highly expressed in colon cancer tissues,and its high expression is a protective factor for the prognosis of COAD patients.
