摘  要:目的探讨穗花杉双黄酮(AMF)对卵巢癌细胞SKOV3迁移能力的影响。方法采用细胞划痕实验与Transwell实验检测AMF对细胞迁移的影响,Western blotting检测AMF对SKOV3细胞黏附相关分子E-钙黏蛋白(E-cadherin)与β-连环蛋白(β-catenin)表达的影响,细胞免疫荧光检测AMF对SKOV3细胞β-catenin表达与分布的影响。结果细胞划痕实验与Transwell实验显示,与对照组相比50μmol·L^(-1)与100μmol·L^(-1)AMF能够显著抑制卵巢癌细胞SKOV3的迁移;Western blotting结果显示,与对照组相比,50μmol·L^(-1)与100μmol·L^(-1)AMF能够显著促进SKOV3细胞E-cadherin与β-catenin的表达;细胞免疫荧光显示,AMF能够显著促进β-catenin的表达,且主要分布于细胞膜。结论AMF能够抑制卵巢癌细胞SKOV3的迁移与侵袭,其主要机制可能与其上调细胞黏附分子E-cadherin/β-catenin复合体表达有关。Objective To investigate the effects of amentoflavone(AMF)on the migration of ovarian cancer cells.Methods Wound healing experiment and Tanswell cell migration assay were adopted to examine the effect of AMF on SKOV3 cell migration.Western blotting was used to analyze the expression level of E-cadherin andβ-catenin.Immunofluorescence test was used to verify the expression and the distribution ofβ-catenin.Results Both 50μmol·L^(-1)and 100μmol·L^(-1)AMF could inhibit the migration of SKOV3 cells as compared with the control group.Western blotting results also showed that 50μmol·L^(-1)AMF and 100μmol·L^(-1)AMF could increase the expression of E-cadherin andβ-catenin in SKOV3 cells.In addition,immunofluorescence test showed thatβ-catenin was mainly located on the cell membrane.Conclusion AMF can inhibit the migration and invasion of ovarian cancer cells through increasing the expression of E-cadherin andβ-catenin proteins.
