摘 要:目的探讨斯钙素(STC)基因在食管癌组织中的表达及临床意义。方法采用GEPIA在线工具分析TCGA数据库食管癌组织中STC-1、STC-2的表达及其与临床预后的关系;免疫组织化学法检测食管癌组织和癌旁组织中STC-2蛋白的表达并分析其临床意义。结果 TCGA数据显示,食管癌组织(n=182)中STC-1、STC-2的mRNA表达水平均显著高于正常组织(n=286),仅STC-2 mRNA的表达与患者总生存期(OS)密切相关(P=0.035)。免疫组化结果显示,80.0%(60/75)的食管癌组织中STC-2蛋白表达水平显著高于配对正常组织(P<0.001),且STC-2蛋白表达与患者18F-氟代脱氧葡萄糖(18F-FDG)的标准化摄取值(SUV)(P<0.001)、临床分期(P=0.033)和OS(P=0.0429)显著相关。结论 STC-2高表达可能在食管癌的发生发展中发挥重要作用,是食管癌预后判断的潜在分子标志物。Objective To explore the expression of stanniocalcin(STC) gene in tissues of esophageal carcinoma(ESCA) patients and its clinical significance. Methods The GEPIA online tool was applied to analyze the mRNA expressions of STC-1 and STC-2 in esophageal cancer tissues of the cancer genome atlas(TCGA) database and their relationship with clinical prognosis. Immunohistochemistry was used to detect the expression of STC-2 protein. Results Based on the TCGA database, the mRNA expression levels of STC-1 and STC-2 were both higher in ESCA tissues(n=182) than in normal tissues(n=286), but only the STC-2 mRNA expression was closely correlated with a poor overall survival(OS)(P=0.035). The immunohistochemical results showed that 60 out of 75 cases(80.0%) had a higher protein expression level of STC-2 in tumor tissues than in adjacent normal tissues(P<0.001). Furthermore, the elevated protein expression of STC-2 was significantly correlated with the maximum standard uptake value(SUV) of 18F-fluorodeoxyglucose(18F-FDG)(P<0.001), advanced clinical stage(P=0.033) and adverse OS(P=0.0429). Conclusion The high expression of STC-2 may play an important role in the development and progression of ESCA, and is a potential biomarker for prognosis assessment of ESCA.
