摘  要:目的研究Pannx1通道阻滞剂对急性癫痫小鼠海马谷氨酸能神经元电生理变化的影响。方法将45只C57 Thy1-YFP小鼠分为癫痫组、Pannx1组和对照组,各15只。Pannx1组在建模前1 h予腹腔注射甲氟喹溶液,癫痫组、Pannx1组建模成功后,3组小鼠直接断头取脑,制作脑片,在人工脑脊液中孵育后,利用膜片钳技术,进行电位记录。观察并比较3组小鼠海马谷氨酸能神经元动作电位峰间距(ISI)和自发兴奋性突触后电流(sEPSC)的间隔和幅度。结果与对照组比较,癫痫组ISI值明显缩短,sEPSC间隔缩短、幅度增大,差异均有统计学意义(均P<0.01)。Pannx1组较癫痫组ISI值延长,sEPSC间隔增大、幅度降低,差异均有统计学意义(均P<0.01)。结论急性癫痫小鼠海马谷氨酸能神经元兴奋性升高;使用甲氟喹后有抗癫痫作用。Objective To investigate the electrophysiological changes of glutaminergic neurons in the hippocampus of mice with acute epilepsy after intervention with Pannx1 channel blocker.Methods Forty-five C57 Thy1-YFP mice were randomly divided into EP group,Pannx1 group and control group with 15 mice each.Epilepsy model was induced by intraperitoneal injection of scopolamine and pilocarpine,and Pannx1 channel blocker mefloquine(MFQ)was intraperitoneally injected in Pannx1 group 1 h before modelling.The brain slices were made and incubated in artificial cerebrospinal fluid,patch-clamp technology was used for whole-cell recording.The inter-spike interval(ISI)and the spacing and amplitude of spontaneous excitatory postsynaptic current(sEPSC)in hippocampal glutamate neurons were compared among the three groups.Results Compared with the control group,the ISI value of epilepsy group was significantly shortened.The SEPSC interval was shortened and the amplitude increased,and the differences were statistically significant(P<0.01).Compared with epilepsy group,the ISI value of Pannx1 group was longer,the SEPSC interval was larger and the amplitude was lower,and the differences were all the same statistical significance(P<0.01).Conclusion The excitability of glutamate neurons in the hippocampus of mice with acute epilepsy was increased,the use of mefloquine has an antiepileptic effect.
