摘  要:目的探讨磷酸二酯酶-5(PDE5)抑制剂对二乙基亚硝胺(DEN)诱导的肝细胞癌(HCC)大鼠的抗肿瘤作用。方法雄性Wistar大鼠分为:对照组、DEN组和PDE5+DEN组,每组10只大鼠。采用免疫印迹法检测各组大鼠肝组织谷胱甘肽S-转移酶(GST)和甲胎蛋白(AFP),并进行肝组织病理学检查。然后测定各组大鼠肝组织匀浆中氧化应激标志物,包括丙二醛(MDA)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GPx)。结果与对照组相比,DEN组大鼠抗氧化酶SOD、CAT和GPx活性降低,同时MDA水平升高(均P<0.05)。与DEN组相比,PDE5+DEN组大鼠抗氧化酶SOD、CAT和GPx活性增加,同时MDA水平降低(均P<0.05)。结论PDE5抑制剂可能通过调节抗氧化途径关键参数来抑制HCC发展,从而发挥抗癌作用。Objective To investigate the anticancer effect of phosphodiesterase-5(PDE5)inhibitor on diethylnitrosamine(DEN)-induced hepatocellular carcinoma(HCC)in rats.Methods Male Wistar rats were divided into control group,DEN group and PDE5+DEN group,with 10 rats in each group.Glutathione S-transferase(GST)and alpha fetoprotein(AFP)in liver tissues of rats in each group were detected by Western blot,and liver histopathological changes were observed.The activities of oxidative stress markers including malondialdehyde(MDA),superoxide dismutase(SOD),catalase(CAT)and glutathione peroxidase(GPx)in liver homogenate of rats in each group were determined.Results Compared with control group,the activities of SOD,CAT and GPX were decreased in DEN group,while the level of MDA was increased(all P<0.05).Compared with DEN group,the activities of SOD,CAT and GPX were increased in PDE5+DEN group,while the level of MDA was decreased(all P<0.05).Conclusion PDE5 inhibitor may prevent the progression of HCC through modulating the key parameters of antioxidant pathway.
