血卟啉单甲醚介导的光动力疗法对胃癌细胞MGC-803的光动力杀伤效应

摘　　要：目的探讨血卟啉单甲醚(HMME)介导光动力疗法(PDT)对胃癌细胞MGC-803的生长抑制作用。方法荧光显微镜观察血卟啉单甲醚在胃癌细胞MGC-803内的分布情况,应用荧光光谱分析仪检测MGC-803细胞内HMME的荧光光谱,从而确定最佳孵育时间。将胃癌细胞MGC-803与不同浓度的HMME孵育90 min后,分别采用不同的光强度照射,使用CCK-8试剂盒分析MGC-803细胞的存活率。结果荧光强度与孵育时间在90 min内呈正相关,在90 min处达到峰值。随着HMME浓度或光照强度的逐渐增加,胃癌细胞MGC-803的生长抑制率显著增加,细胞存活率与HMME浓度、光强度的乘积呈e指数衰减,光动力因子B=光输出功率×照射时间×HMME浓度(B=I_(0)×t×C)。结论HMME介导的PDT可显著抑制HMC-803细胞的增殖,光动力因子B可为临床的个体化治疗提供依据。Objective To explore the inhibitory effect of hematoporphyrin monomethionine(HMME)-mediated photodynamic therapy(PDT)on the growth of gastric cancer cell MGC-803.Methods Fluorescence microscope was used to observe the distribution of hematoporphyrin monomethionine in gastric cancer cells MGC-803,and the fluorescence spectrum of HMME in MGC-803 cells was detected with a fluorescence spectrum analyzer to determine the best incubation time.After incubating gastric cancer cells MGC-803 with different concentrations of HMME for 90 min,they were irradiated with different light intensities,and the survival rate of MGC-803 cells was analyzed using the CCK-8 kit.Results The fluorescence intensity was positively correlated with the incubation time within 90 mins,and reached a peak at 90 th.With the gradual increase of HMME concentration or light intensity,the growth inhibition rate of gastric cancer cell MGC-803 increases significandy.The product of cell survival rate,HMME concentration and light intensity decays exponentially,photodynamic factor B=light output power×irradiation time×HMME concentration(B=I_(0)×t×C).Conclusion HMME-mediated PDT can significantly inhibit the proliferation of HMC-803 cells,and photodynamic factor B can provide a basis for clinical individualized treatment.
