摘  要:目的探讨艾塞那肽对人卵巢癌OVCAR-3细胞凋亡、侵袭和迁移的影响及机制。方法选取人卵巢癌OVCAR-3细胞作为研究对象,并分为艾塞那肽组和对照组,艾塞那肽组给予对数生长期的人卵巢癌OVCAR-3细胞10-100μM的艾塞那肽处理,对照组不给予艾塞那肽处理。采用CCK-8法检测细胞活力,并计算其半数抑制浓度(IC50)。在确定的IC50浓度基础上,采用Transwell实验检测OVCAR-3细胞侵袭和迁移能力,流式细胞术检测细胞周期,实时定量聚合酶链式反应(PCR)法检测凋亡相关蛋白Bax和Bcl-2的表达情况,免疫印迹法检测核转录因子kappa-B(NF-κB)的表达及其乙酰化水平。结果 CCK-8试验结果显示,艾塞那肽对OVCAR-3细胞的半数抑制浓度(IC50)为40.08μM。与对照组相比,艾塞那肽可使人卵巢癌OVCAR-3细胞凋亡和细胞凋亡相关蛋白Bax的表达水平升高,细胞侵袭数、细胞迁移数、Bcl-2、NF-κB以及乙酰化NF-κB(acyl-NF-κB)的表达降低,差异均有统计学意义(均P<0.05)。结论艾塞那肽可抑制人卵巢癌OVCAR-3细胞的活力,促进其凋亡并抑制其侵袭和迁移能力,其作用机制可能与抑制NF-κB及其乙酰化表达有关。Objective To investigate the effect of exenatide on the apoptosis,invasion and migration of OVCAR-3 human ovarian cancer cells line. Methods The OVCAR-3 human ovarian cancer cells were selected and divided into an exenaptide group and a control group. For the exenaptide group,human ovarian cancer OVCAR-3 cells was given 10 to 100 μm exenatide during logarithmic growth phase,while the control group was not processed with exenatide. Cell viability was detected by CCK-8 kit. Half inhibit concentration( IC50) was calculated. Based on the determined IC50 concentration,the invasion and migration of OVCAR-3 cells were detected using Transwell test. Cell cycle was detected by flow cytometry. The expression of apoptosis-related proteins Bax and Bcl-2 was detected by real-time quantitative PCR. The expression and acetylation levels of NF-κB were detected by western blotting. Results CCK-8 test showed that the IC50 of exenatide for OVCAR-3 cells was 40. 08 μM. Compared with the control group,exenatide significantly increased the apoptosis of human ovarian cancer OVCAR-3 cells,the expression of Bax,the number of invasive cells,the number of cell migration,and the expression of Bcl-2,NF-κB and acetylated NF-κB( acyl NF-κB)( all P<0.05). Conclusion Exanaptide can inhibit the activity of OVCAR-3 human ovarian cancer cells,promote the apoptosis,and inhibit the invasion and migration. The mechanism of action may be related to the inhibition of NF-κB and its acetylation expression.
