摘  要:目的检测新型金雀异黄素衍生物HNPG对骨肉瘤MG-63细胞迁移和侵袭的抑制作用及其可能分子生物学机制。方法体外培养骨肉瘤MG-63细胞,用MTT检测HNPG对MG-63细胞的增殖抑制作用;用划痕实验检测HNPG对MG-63细胞迁移抑制作用;用Transwell检测HNPG对MG-63细胞侵袭的抑制作用;用PI染色FCM检测HNPG对MG-63细胞G1期的阻滞作用;用AV-PI染色FCM检测HNPG对MG-63细胞凋亡的诱导作用;用Western blotting检测HNPG对MG-63细胞p53、bcl-2和bax蛋白表达的调节作用。结果HNPG在体外呈时间和浓度依赖性抑制MG-63细胞增殖,24 h的IC50为(4.8±0.6)μmol/L,5μmol/L的HNPG作用MG-63细胞24 h增殖抑制率为(45.2±4.6)%,较NS组的(0.40±0.02)%显著升高,差异有统计学意义(P<0.05),划痕愈合率为(38.24±2.48)%,较NS组的(71.23±4.62)%显著降低,差异有统计学意义(P<0.05),侵袭抑制率为(44.66±2.06)%,较NS组的(0.34±0.03)%显著升高,差异有统计学意义(P<0.05),伴随着细胞G1期比例增高(78.65±3.48)%,较NS组的(64.40±2.02)%显著升高,差异有统计学意义(P<0.05),细胞凋亡率增高(24.63±2.12)%,较NS组的(2.75±0.20)%显著升高,差异有统计学意义(P<0.05);同时p53和bax蛋白表达上调,而bcl-2蛋白表达下调,与NS组比较,差异有统计学意义(P<0.05)。结论HNPG在体外可以显著抑制MG-63细胞增殖、迁移和侵袭,其机制可能与其阻滞细胞于G1期、上调p53蛋白表达,进而激活bax蛋白表达、抑制bcl-2蛋白表达、诱导细胞凋亡所致。Objective To detect the inhibitory effect of novel genistein derivatives HNPG on the migration and invasion of osteosarcoma MG-63 cells and its possible molecular biological mechanism.Methods Osteosarcoma MG-63 cells were cultured in vitro,and MTT was used to detect the proliferation inhibitory effect of HNPG on MG-63 cells.The scratch test was used to detect the migration inhibitory effect of HNPG on MG-63 cells.The Transwell was used to detect the inhibitory effect of the invasion of HNPG on MG-63 cells.PI staining FCM was used to detect the blocking effect of HNPG on the G1 phase of MG-63 cells.AV-PI staining FCM was used to detect the apoptosis induction effect of HNPG on MG-63 cells.Western blotting was used to detect the regulatory effect of HNPG on the expression of p53,bcl-2,and bax proteins in MG-63 cells.Results HNPG inhibited the proliferation of MG-63 cells in a time and concentration-dependent manner in vitro.The IC50 of 24 h was(4.8±0.6)μmol/L.The 24 h proliferation inhibition rate of MG-63 cells treated by 5μmol/L of HNPG was(45.2±4.6)%,which was higher than that of(0.40±0.02)%in the NS group,the difference was significant(P<0.05).The scratch healing rate was(38.24±2.48)%,which was lower than that of(71.23±4.62)%in the NS group,the difference was significant(P<0.05).The invasion inhibition rate was(44.66±2.06)%,which was higher than that of(0.34±0.03)%in the NS group,the difference was significant(P<0.05).The proportion of cells in the G1 phase(78.65±3.48)%,which was higher than that of(64.40±2.02)%in the NS group,the difference was significant(P<0.05).The apoptosis rate was increased more statistically(24.63±2.12)%,which was higher than that of(2.75±0.20)%in the NS group,the difference was significant(P<0.05).At the same time the expression of p53 and bax protein was up-regulated,while the expression of bcl-2 protein was down-regulated,which was statistically significant compared with the NS group(P<0.05).Conclusion HNPG can significantly inhibit the proliferation,migration,and invasi
