摘　　要：目的探讨IL-23受体(IL-23R) rs10889677位点单核苷酸多态性与大肠癌易感性的关系。方法收集21例大肠癌患者(A组)癌组织及20例肠镜检查正常患者(B组)的正常大肠黏膜标本,并抽取外周血,采用高分辨率熔解曲线分析法比较两组IL-23R rs10889677位点基因型分布和等位基因频率。结果两组外周血及组织IL-23R rs10889677位点基因型分布及等位基因频率均有统计学差异(P<0.05)。IL-23R rs10889677位点基因型分布在两组中均符合Hardy-Weinberg平衡(P>0.05)。多因素logistic回归分析显示,在校正患者年龄、性别、吸烟史、饮酒史和部位等之后,外周血和组织中IL-23R rs10889677位点携带CC基因型的患癌风险分别是携带AA基因型的12.630倍和72.938倍(P<0.05)。结论 IL-23R rs10889677位点单核苷酸多态性可能与大肠癌遗传易感有关。Objective To explore the relationship between IL-23 R rs10889677 single nucleotide polymorphism and colorectal cancer susceptibility.Methods The cancer tissues of 21 patients with colorectal cancer(group A)and the normal colorectal mucosal specimens of 20 patients with normal colonoscopy(group B)were collected and peripheral blood was drawn.The high-resolution melting curve analysis method was used to analyze the genotype distribution and allele frequency of IL-23 R rs10889677 of the two groups.Results There were significant differences in the genotype distribution and allele frequency of IL-23 R rs10889677 in peripheral blood and tissues between the two groups(P<0.05).The genotype distribution of IL-23 R rs10889677 locus in two groups accorded with Hardy-Weinberg equilibrium(P>0.05).Multivariate logistic regression analysis showed that after adjusting for age,gender,smoking history,drinking history and location,the risk for colorectal cancer with CC genotype at IL-23 R rs10889677 in peripheral blood and tissues were 12.630 times and 72.938 times higher than that with AA genotype(P<0.05).Conclusion The single nucleotide polymorphism of IL-23 R rs10889677 may be related to the genetic susceptibility of colorectal cancer.
