摘  要:目的对尼古丁诱导癫痫发作新皮质中发现的差异表达基因(DEGs)进行生物信息学分析。方法利用DAVID 6.8和STRING 11.0等分析平台对基因芯片公共数据库(GEO)发表的尼古丁诱导癫痫发作的新皮质筛选DEGs,并进行基因本体(GO)功能富集分析、京都基因与基因组百科全书(KEGG)信号通路分析和蛋白质相互作用网络构建。结果癫痫发作新皮质中发现的DEGs与癫痫致病性基因构建网络,结果共筛选出95个上调基因及119个下调基因,DEGs主要富集于胞嘧啶上的DNA甲基化、DNA复制偶联的核小体组装、细胞凋亡过程的调控等生物学过程,以及刺激神经组织的配体受体相互作用通路、核转录因子(NF-κB)信号通路等。结论尼古丁诱导癫痫发作新皮质相比正常新皮质的DEGs存在明显差异。多个信号通路、多个蛋白质相互作用方式为理解癫痫发作新皮质病理过程提供了帮助。Objective To analyze the bioinformatics of differentially expressed genes in the the neocortex of epileptic seizures induced by nicotine.Methods Neocortex differentially expressed genes of nicotine-induced epileptic seizures published in GEO were screened by DAVID 6.8,and STRING 11.0.The gene ontology(GO)function enrichment analysis,Kyoto Encyclopedia of Genes and Genomes(KEGG),and protein interaction network construction were performed.Results A total of 95 up-regulated and 119 down-regulated genes were selected,and the differentially expressed genes were mainly enriched in cytosine DNA methylation,DNA replication coupled nucleosome assembly,cell apoptosis regulation and other biological processes,and ligand receptor interaction pathway and NF-kappa B signaling pathway that stimulate nerve tissue.Conclusion There are significant differences in the expression of nicotine-induced epileptic neocortex compared with normal neocortex.Multiple signal pat hways and multiple protein interaction patterns are helpful to understand the neocortical pathological process of epileptic seizures.
