河北省首例D8基因亚型麻疹病毒株的分子流行病学分析

摘  要:目的开展麻疹患者的病原学和分子监测,加强麻疹病毒基因型的监测。方法利用Vero/SLAM细胞(淋巴信号激活因子转染的非洲绿猴肾细胞),按常规方法分离病毒;利用RT-PCR扩增麻疹病毒N基因,然后测序并构建基因亲缘性关系树。对麻疹疫苗株S191和此次分离的D8野毒株进行抗体中和试验,计算中和抗体滴度,比较疫苗对不同毒株的保护效果。结果该麻疹病毒分离株与世界卫生组织(WHO)D8基因亚型参考株在基因亲缘性关系树上同属1个分支,核苷酸和氨基酸同源性分别为98.7%和97.3%;与2019年第44周我国香港监测到的D8基因亚型麻疹病毒流行株的核苷酸和氨基酸相似性均为100%。所选血清标本对麻疹疫苗株和此次分离的D8野毒株的中和抗体几何平均滴度分别为1∶50.80和1∶40.32,中和抗体滴度无明显差异。结论河北省首次监测到输入型D8基因亚型麻疹患者。Objective To conduct the etiology and molecular surveillance on patients with mealsles,and enhance the surveillance on genotypes of measles virus.Methods The measles viruses were isolated with Vero/SLAM cell(African green monkey kidney cells transfected with lymphoid signal activating factor)by routine methods.Nucleoprotein(N)genes of measles viruses were amplified by RT-PCR,the sequencing was made,and the gene phylogenetic tree was constructed.Measles Vaccine Strain S191 and currently isolated D8 wild strains were subject to antibody neutralization test,the neutralization antibody titer were calculated,and the protective effects of the vaccine on different virulent strains were compared.Results The measles virus isolate and WHO D8 gene subtype reference strain shared the same branch on the gene phylogenetic tree,the homology of nucleotide acids and amino acids was 98.7%and 97.3%respectively.The similarities of nucleotide acids and amino acids of epidemic strains of D8 gene subtype measles virus detected in Hong Kong in the 44 th week,2019 were both 100%.The geometric mean neutralizing antibody titer of selected serum specimen against measles vaccine strain and D8 wild strain isolated currently was 1∶50.80 and 1∶40.32 respectively,and there was no significant difference in terms of neutralization antibody titer.Conclusion The patient with imported D8 gene subtype measles was detected for the first time in Hebei Province.
