摘 要:目的观察组织工程肌腱复合鹿茸多肽修复大鼠髌韧带缺损的效果,探讨促进肌腱损伤修复的有效方法。方法将36只SD大鼠随机分为3组,每组12只。对3组大鼠髌韧带做1 mm×4 mm全层窗口缺损,模型组缺损待自然修复,组织工程组采用组织工程肌腱(采用大鼠肌腱细胞构建)修复,组织工程复合鹿茸多肽组采用组织工程肌腱复合鹿茸多肽修复。4周后,检测修复组织的外观形态、组织学结构和胶原含量。结果模型组外观有皱缩现象,生成较多瘢痕组织,胶原含量最少;组织工程组外观较平整,组织结构规则,胶原含量居中;组织工程复合鹿茸多肽组外观平整光滑,修复组织内部结构规则良好,胶原含量最高。3组间胶原含量比较差异均有统计学意义(P均<0.05)。结论组织工程肌腱有助于大鼠髌韧带缺损的修复,组织工程肌腱联合鹿茸多肽修复作用更佳,鹿茸多肽促进大鼠髌韧带缺损修复的原因可能与促进肌腱细胞分泌胶原蛋白有关。Objective It is to observe the effect of tissue engineered tendon combined with velvet antler polypeptide in the repair of rat patellar ligament defect,and to explore the effective methods to promote the repair of tendon injury.Methods Thirty-six SD rats were randomly divided into 3 groups,each group had 12 rats.A 1 mm×4 mm full-thickness window defect was performed on the patellar ligament of the rats of the 3 groups.The defect in the model group was to be repaired naturally,the tissue engineering group was repaired with tissue engineered tendon(built with rat tendon cells),and the tissue engineering combined with deer antler polypeptide group(combination group)was repaired with tissue engineered tendon combined with deer antler polypeptide.After 4 weeks,the appearance,histological structure and collagen content of the repaired tissue were examined.Results The appearance of the model group was shrinking,more scar tissue was generated,and the collagen content was the least;the tissue engineering group had a smoother appearance,regular tissue structure,and the collagen content was in the middle;the combination group had a smooth appearance,and the internal structure of the repair tissue was regular,the collagen content was the highest.The differences in collagen content among the three groups were statistically significant(all P<0.05).Conclusion Tissue engineered tendon is helpful for the repair of rat patellar ligament defect.Tissue engineered tendon combined with velvet antler polypeptide has a better repair effect.The reason why velvet antler polypeptide promotes the repair of rat patellar ligament defect may be related to the promotion of collagen secretion by tendon cells.
