摘 要:目的:探讨益气活血方、方中益气药和活血药对气虚血瘀模型大鼠气血的影响。方法:采用荧光微球致多发性脑梗复合睡眠剥夺法制备气虚血瘀型脑梗的病证结合模型,将大鼠随机分为假手术组、模型组、益气活血方组(3.49 g·kg^(-1))、益气药组(1.08 g·kg^(-1))、活血药组(2.408 g·kg^(-1)),脑梗术后复合睡眠剥夺4周,期间每日给药1次。给药4周后测定各组大鼠的负重力竭游泳时间和脉搏幅度,取血进行脑组织血液相关指标及病理切片分析。结果:相较假手术组,模型组动物负重力竭游泳时间明显缩短,脉搏幅度也明显下降(P<0.01);模型组在各切变率下的全血黏度和血浆黏度相比假手术组均显著升高(P<0.01),血清中ET-1、Ang-Ⅱ、IL-6、IL-1β和MDA水平均升高(P<0.05)。与模型组比较,益气活血方组能延长负重游泳时间,提高脉搏幅度(P<0.05);活血药能降低各切变率下的全血黏度,益气药和全方可降低血浆黏度(P<0.05);益气药能降低Ang-Ⅱ值,益气组与活血组对ET-1都有不同程度的降低(P<0.05),全方优于益气药和活血药组(P<0.01);各给药组IL-1β值相比模型组均有显著降低,活血药组IL-6也有明显降低(P<0.01);活血药和全方MDA水平低于模型组(P<0.05,P<0.01)。病理形态学观察表明,模型组海马组织结构异常,见坏死和炎性浸润,益气组见神经元增生,活血组未见坏死区,全方组病理改变最少,且见炎性组织中细胞恢复状态。结论:益气药与活血药存在协同作用,益气以助血行,改善气虚血瘀,同时活血化瘀进而减少炎症因子及氧化产物的堆积,使全方效果既优于益气药也优于活血药。Objective:To study the effects of Yiqi Huoxue prescription,Yiqi and Huoxue medicine on rats with Qi-deficiency and blood stasis.Methods An animal model of combination of disease and syndrome was established by microspheres and sleep deprivation induced cerebral ischemia with Qi deficiency and blood stasis.The model rats were randomly divided into model group,Yiqi Huoxue prescription group,Yiqi medicine group,and Huoxue medicine group(3.49 g·kg^(-1)、1.08 g·kg^(-1)、2.408 g·kg^(-1)).After the operation of multiple cerebral infarction caused by fluorescent microspheres,the rats were given sleep deprivation for 4 weeks,once a day.The rats in the sham group were only separated blood vessels without sleep deprivation.Before the end of administration,the exhaustive weight-bearing swimming time and pulse amplitude of rats in each group were measured.The plasma and whole blood rheological indexes were detected;the contents of interleukin-6(IL-6),interleukin-1β(IL-1β),endothelin-1(ET-1),angiotensin II(Ang-II)and malondialdehyde(MDA)in serum of each group were detected;histopathological changes of rat brain were observed by hematoxylin-eosin staining.Compared with the shame group,the exhaustive weight-bearing swimming time of animals in the model group was significantly shorter and the pulse amplitude was also significantly decreased(P<0.01);the whole blood viscosity and plasma viscosity of the model group a teach shear rate were significantly increased(P<0.01),and the serum ET-1,Ang-II,IL-6,IL-1βand MDA levels were increased(P<0.05).Results Compared with the model group,the Yiqi Huoxue prescription could prolong the exhaustive weight-bearing swimming time and increase the pulse amplitude(P<0.05);Huoxue medicine could reduce the whole blood viscosity a teach share rate,and Yiqi medicine and the whole prescription could reduce the plasma viscosity(P<0.05);Yiqi medicine could reduce the Ang-II value in serum,the animals in both Yiqi medicine group and Huoxue medicine group had different degrees of reduction in ET-1(P
