黄连素联合miR-19b-3p调控阿尔茨海默病的实验研究

摘　　要：目的探讨黄连素联合miR-19b-3p对阿尔茨海默模型细胞发生发展的影响。方法实验设置对照(NC)组、Aβ组、0.1μmol/L黄连素+Aβ组、1.0μmol/L黄连素+Aβ(黄连素+Aβ)组、10.0μmol/L黄连素+Aβ组、miR-NC+Aβ组、miR-19b-3p+Aβ组、黄连素+miR-NC+Aβ组、黄连素+miR-19b-3p+Aβ组。细胞计数试剂盒8(CCK-8)检测细胞增殖;蛋白质印迹(Western Blot)法检测细胞周期蛋白D1(Cyclin D1)、裂解的半胱氨酸天冬氨酸蛋白酶-3(Cleaved-caspase-3)蛋白表达;流式细胞术检测细胞凋亡;实时荧光定量PCR(RT-qPCR)检测miR-19b-3p表达水平。结果Aβ诱导的PC12细胞中CyclinD1表达水平显著降低,Cleaved-caspase-3表达水平显著升高,细胞OD值显著降低,细胞凋亡率显著升高,miR-19b-3p表达水平显著降低(P<0.05)。黄连素或过表达miR-19b-3p可促进Aβ作用的PC12细胞增殖,抑制细胞凋亡。黄连素联合miR-19b-3p可更显著的抑制Aβ作用的PC12细胞凋亡。结论黄连素联合miR-19b-3p减轻了Aβ诱导的PC12细胞损伤,对阿尔茨海默病模型细胞损伤具有保护作用。Objective To investigate the effects of berberine combined with miR-19b-3p on the occurrence and development of Alzheimer’s disease model cells.Methods The experiment was divided into control group,Aβgroup,0.1μmol/L berberine+Aβgroup,1.0μmol/L berberine+Aβ(berberine+Aβ)group,10.0μmol/L berberine+Aβgroup,miR-NC+Aβgroup,miR-19b-3p+Aβgroup,berberine+miR-NC+Aβgroup,berberine+miR-19b-3p+Aβgroup.Cell counting kit 8(CCK-8)was used to detect the cell proliferation;Western Blot was used to detect the expressions of cyclin D1(Cyclin D1)and cleaved cysteine aspartic protease-3(Cleaved-caspase-3)protein;flow cytometry was used to detect cell apoptosis;real-time fluorescence quantitative PCR(RT-qPCR)was used to detect the expression of miR-19b-3p.Results The expression levels of CyclinD1 and miR-19b-3p as well as cell OD value were significantly decreased in Aβ-induced PC12 cells,however,the expression levels of Cleaved-caspase-3 and cell apoptotic rate were significantly increased(P<0.05).Berberin or overexpression of miR-19b-3p could promote the proliferation of PC12 cells treated by Aβand inhibit cell apoptosis.Moreover Berberin combined with miR-19b-3p could significantly inhibit the apoptosis of PC12 cells treated by Aβ.Conclusion Berberin combined with miR-19b-3p can relieve PC12 cell damage induced by Aβand has protective effects on Alzheimer’s disease model cells.
