摘  要:目的探讨miR-209下调MEK/ERK信号通路对胶质瘤U87细胞增殖、增强放疗敏感性的影响。方法采用Real Time PCR的方法检测胶质瘤U87细胞放疗前后miR-209、MEK、ERK mRNA的表达;通过转染技术使U87细胞内过表达miR-209,Western blot、Real time PCR检测细胞内MEK、ERK表达水平。结果放疗后胶质瘤U87细胞中miR-209相对表达降低(t=2.491,P=0.037 4),放疗前后ERK、ERKmRNA的相对表达差异无统计学意义;miR-209 mimics转染U87细胞后,MEK mRNA表达减少(0.11±0.05),ERK mRNA表达减少(F=9.891,P=0.002 9),p-MEK、p-ERK表达均降低。miR-209转染后24、48 h,细胞增殖受到抑制。结论 miR-209通过下调胶质瘤U87细胞中MEK-ERK的表达,能抑制细胞增殖,增强放疗敏感性。Objective To investigate the effect of miR-209 down regulating the MEK/ERK signal pathway on the proliferation and radiosensitivity of glioma U87 cells.Methods The expressions of miR-209,MEK and ERK mRNA in U87 cells before and after radiotherapy were detected by Real Time PCR.After miR-209 transfection,the expressions of miR-209,ERK and MEK were detected by Western blot and real time PCR.Results After radiotherapy,the relative expression of miR-209 in U87 cells decreased(t=2.491,P=0.0374),and there was no significant difference in ERK and ERK mRNA expressions between pre-and post-radiotherapy.After the miR-209was transfected to the U87 cells,the MEK mRNA expression decreased(0.11±0.05),The ERK mRNA expression decreased(F=9.891,P=0.0029),and the p-MEK and p-ERK expressions both decreased,and cell proliferation was inhibited.Conclusion miR-209 can inhibit cell proliferation and enhance radiosensitivity by down regulating the expression of MEK-ERK in U87 cells.
