摘 要:青杨天牛(Saperda populnea)是一种主要以北京杨、新疆杨和藏川杨等绿化树种为食的蛀干害虫,其虫害在西藏每年爆发一次,对城市生态景观造成严重影响。本研究利用高通量测序方法,以青杨天牛肠道细菌为研究对象,分析了拉萨市3个区域青杨天牛肠道细菌多样性。结果表明,青杨天牛肠道细菌Shannon多样性排序为:HBL>JYC>LBLK;其肠道细菌优势菌门为:Proteobacteria、Actinobacteria、Bacteroidetes、Verrucomicrobi、Firmicutes、Cyanobacteria和Chloroflexi;其优势菌属为Wolbachia、Raoultella、Pantoea;3个区域青杨天牛肠道细菌在Allobaculum、Erwinia、Serratia、Marmoricola和RB-25-group等菌属存在显著差异(0.01<P<0.05);3个区域青杨天牛肠道细菌共享菌属约为68.57%,其中关于纤维素分解的细菌在3个区域的青杨天牛肠道中共享程度较高,且整体菌群以正相关为主。通过探究青杨天牛肠道细菌结构组成和多样性特征,为青杨天牛的生物防治提供了数据支撑。Saperda populnea is a kind of Trunk borers that mainly feeds on Populus beijingensis,Populus bolleana Lauche and Populus pamirica var.tibetica.The Trunk borer pest outbreaks once a year in Tibet,causing serious impacts on the urban ecological landscape.In this study,high-throughput sequencing was used to analyze the diversity of intestinal bacteria in Saperda populnea from three areas of Lhasa City.The results showed that the Shannon-Wiener diversity Index of intestinal bacteria was HBL>JYC>LBLK;The dominant phyla of intestinal bacteria are Proteobacteria,Actinobacteria,Bacteroidetes,Verrucomicrobi,Firmicutes,Cyanobacteria and Chloroflexi.The dominant Genus were Wolbachia,Raoultella and Pantoea.There were significant differences in Allobaculum,Erwinia,Serratia,Marmoricola and RB-25-group of intestinal bacteria among the three regions(0.01<P<0.05).About 68.57%of Saperda populnea intestinal bacteria were shared bacteria.Among them,bacteria related to cellulose decomposition were shared to a higher degree in intestinal bacteria of Saperda populnea from three regions,and the overall flora was mainly positively correlated.By exploring the structural composition and diversity characteristics of intestinal bacteria in Saperda populnea,it provides data support for biological control of Saperda populnea.
