摘  要:目的观察不同浓度溴芬酸钠对紫外线诱导的人晶状体上皮细胞损伤的修复作用,比较前列腺素E2及环氧化酶-1,环氧化酶-2的mRNA以及蛋白表达变化。方法采用不同时长的紫外线照射处理人晶状体上皮细胞,MTT法检测细胞活性变化;UV照射30s,溴芬酸钠(80μg/ml)作用紫外线诱导的人晶状体上皮细胞,MTT法检测细胞活性的改变,ELISA法检测细胞上清液中前列腺素E2的浓度,使用RT-qPCR检测COX-1、COX-2 mRNA的表达,Western blot检测COX-1、COX-2蛋白表达量。结果随着紫外线照射时间的延长,人晶状体上皮细胞活性下降,上清液中前列腺素E2的浓度增加。溴芬酸钠处理后,细胞活性显著上升,上清液中前列腺素E2的生成下降,抑制前列腺素E2生成的程度与溴芬酸钠浓度呈正相关。紫外线照射可引起人晶状体上皮细胞中COX-2 mRNA及蛋白的增加,溴芬酸钠选择性抑制COX-2 mRNA及蛋白的生成。结论溴芬酸钠抑制紫外线对人晶状体上皮细胞的氧化损伤,与选择性抑制环氧化酶-2和减少前列腺素E2生成相关。Objective To observe the different concentrations of bromfenate sodium on the expression of prostaglandin E2 and cyclooxygenase-1 and cyclooxygenase-2 mRNA and protein in human lens epithelial cells induced by ultraviolet(UV). Methods The human lens epithelial cells SRA01/04 was irradiated with UVB(0, 10, 20, 30, 40 s), the human lens epithelial cells induced by UVB(30 s) were treated with sodium bromfenate(80μg/ml), the cell viability was detected by MTT, the concentration of prostaglandin E2 in cell supernatant was detected by ELISA, the expression of COX-1 and COX-2 mRNA in ultraviolet induced HLECs was detected by qRT-PCR, and the expression of COX-1 and COX-2 protein was detected by Western blot. Results The activity of human lens epithelial cells was decreased and the concentration of prostaglandin E2 in supernatant was increased, with the prolongation of ultraviolet irradiation. Bromfenate sodium up-regulated the cell activity, reduced the production of prostaglandin E2 in the supernatant with dose-dependent manner. Ultraviolet irradiation increased the expression level of COX-2 mRNA and protein in human lens epithelial cells, however, Bromfenate sodium selectively inhibited the production of COX-2 mRNA and protein. Conclusion The inhibitive effect of Bromfenate sodium on the oxidative damage of human lens epithelial cells caused by ultraviolet is related to inhibition of cyclooxygenase-2 and prostaglandin E2.
