摘　　要：目的探讨罂粟碱对非小细胞肺癌A549和H460细胞增殖、凋亡和迁移能力的影响,并初步探讨其分子机制。方法利用罂粟碱处理非小细胞肺癌A549和H460细胞,CCK-8细胞增殖检测试剂盒和平板克隆实验分析罂粟碱对A549和H460细胞增殖能力的影响,流式细胞术检测罂粟碱对A549和H460细胞凋亡的影响,Transwell小室实验检测罂粟碱对A549和H460细胞迁移能力的影响;Western blot分析罂粟碱对A549和H460细胞HMGB1、NLRP3、caspase-1、IL-1β和IL-18蛋白表达的影响。结果与对照组相比,经罂粟碱处理后,A549和H460细胞增殖和迁移能力显著下降,凋亡率明显上升,HMGB1、NLRP3、caspase-1、IL-1β和IL-18蛋白表达水平均显著降低。结论罂粟碱下调HMGB1蛋白表达,影响炎症小体活性,抑制A549和H460细胞增殖和迁移,并诱导凋亡。Objective To investigate the effect of Papaverine on the proliferation, apoptosis and migration in non-small-cell lung cancer A549 and H460 cells, to explore its molecular mechanism. Methods The cell proliferation ability was detected by CCK-8 kit and plate clone formation assay, the apoptosis was detected by flow cytometry, and the cell migration ability was detected by transwell. Western blot was used to analyze the expression of HMGB1, NLRP3, caspase-1, IL-1β and IL-18 in non-small-cell lung cancer A549 and H460 cells. Results Compared with control group, Papaverine significantly reduced A549 and H460 cells proliferation ability and migration ability, raised apoptosis rate, decreased HMGB1, NLRP3, caspase-1, IL-1βand IL-18 proteins expression levels. Conclusion Papaverine effectively inhibits A549 and H460 cells proliferation, and migration, promotes apoptosis via inhibiting HMGB1 mediated NLRP3 inflammasome.
