摘  要:目的:运用代谢组学方法考察冰水浴对正常生理状态下大鼠肝脏内源性物质能量代谢的影响,寻找相关潜在生物标志物并分析其能量代谢途径,探讨其机制。方法:将SD雄性大鼠随机分为空白组和冰水浴诱导的寒凝血瘀大鼠模型组,对两组大鼠细胞色素C氧化酶(COX)、ATP合酶(ATPase)、腺苷激酶(ADK)、Na^(+)-K^(+)-ATP酶的能量代谢相关酶进行检测,应用UPLC-Q-TOF-MS技术对空白组和模型组大鼠肝脏进行分析,探求其潜在的生物标志物及相关性最强的物质能量代谢通路。结果:与空白组相比,模型组大鼠肝脏中COX和Na^(+)-K^(+)-ATP酶的含量显著性降低(P<0.05),ATPase和ADK的含量极显著性降低(P<0.01)。同时找出与模型大鼠比较具有显著差异的生物标志物25个,涉及8条相关的代谢通路,其中相关性最强的代谢通路为甘油磷脂代谢(Glycerophospholipid metabolism)。结论:推测冰水浴通过影响正常大鼠的能量代谢阶段相关指标、肝脏代谢产物和能量代谢相关潜在代谢通路,抑制机体能量代谢和功能活动,符合寒证特点。Objective:To explore the effect of ice-water bath on energy metabolism of endogenous substances in liver of rats by using metabolomics method,and to find potential biomarkers and analyze their metabolic pathways to clarify the action mechanism.Methods:The rats model of cold coagulation and blood stasis syndrome was induced by ice-water bath.The contents of Cytochrome C oxidase(COX),ATP synthetase(ATPase),Adenosine kinase(ADK)and Na^(+)-K^(+)-ATPase were detected by enzyme-linked immunosorbent assay.UPLC-Q-TOF-MS technique was used to explore the potential key biomarkers and their related metabolic pathways.Results:Compared to those in the blank group,the contents of COX and Na^(+)-K^(+)-ATPase in liver were significantly decreased(P<0.05),and the contents of ATPase and ADK were extremely reduced in the model group(P<0.01).A total of 25 biomarkers different from the model rats were identified,involving 8 related metabolic pathways,of which glycerophospholipid metabolism was the most relevant.Conclusion:It is suggested that ice-water bath inhibit energy metabolism and functional activities by influencing relevant indexes of energy metabolism,liver metabolites and related potential metabolic pathways,which met the characteristics of cold syndrome.
