摘  要:探讨长链非编码RNA(lncRNA)DSCAM-AS1对子宫内膜癌(EC)发生发展的影响和机制。研究发现与癌旁组织比较,EC组织中DSCAM-AS1表达升高,微RNA-299-3p(miR-299-3p)表达降低,且EC组织中DSCAM-AS1和miR-299-3p的表达呈负相关。DSCAM-AS1在Ishikawa细胞中负调控miR-299-3p表达。干扰DSCAM-AS1后,Ishikawa细胞OD值、克隆形成数、迁移和侵袭数降低;而干扰miR-299-3p降低了干扰DSCAM-AS1对Ishikawa细胞增殖、迁移和侵袭的影响。DSCAM-AS1可能通过竞争性结合miR-299-3p促进EC细胞增殖、迁移和侵袭。The purpose of this study was to investigate the effect and mechanism of the long-chain non-coding RNA(lncRNA)DSCAM-AS1 on development and progression of endometrial carcinoma(EC).The study found that compared with adjacent tissues,the EC tissues showed upregulated expression of DSCAM-AS1 and down-regulated expression of microRNA-299-3p(miR-299-3p).In EC tissues,the expression of DSCAM-AS1 was negatively correlated with that of miR-299-3p.DSCAM-AS1 was shown to negatively regulate the expression of miR-299-3p in Ishikawa cells.After interfering the DSCAM-AS1,the OD value,number of clone formation,number of migratory and invasive Ishikawa cells were lowered.Interference with miR-299-3p reduced the effect of DSCAM-AS1 interference on the proliferation,migration and invasion of Ishikawa cells.DSCAM-AS1 might promote the proliferation,migration and invasion of EC cells by competitively binding to miR-299-3p.
