摘  要:CD47是一种新的免疫抑制信号分子,其与子宫内膜癌进展和预后的关系尚不清楚。通过综合分析TCGA、GEO和THPA数据库中子宫内膜癌的基因和蛋白表达数据及临床资料,发现:①CD47在子宫内膜癌中表达升高并与不良预后相关;②对CD47及其他差异表达基因所在的GO通路进行富集分析,发现CD47相关的GO通路主要为调控肌动蛋白细胞骨架构建过程的通路,该通路相关基因包括FAM49B、CLIP2、DIAPH1和F2RL1的表达升高与CD47表达升高有关,而它们的表达升高又与子宫内膜癌不良预后相关;③CD47及FAM49B、CLIP2在高级别的子宫内膜癌中表达明显升高。以上结果表明,CD47及其相关的调控肌动蛋白细胞骨架通路在子宫内膜癌的进展中发挥重要作用,CD47可能是判断子宫内膜癌预后的有用生物标志和潜在的免疫治疗靶点。CD47,a novel immunosuppressive signal molecule,its relationship with the progression and prognosis of endometrial cancer is ill defined.Through a comprehensive analysis of the gene and protein expression data and clinical data of endometrial cancer in the TCGA,GEO and THPA databases,we found that:①CD47 expression was elevated in endometrial cancer and was associated with a poor prognosis;②The enrichment analysis on GO pathways of CD47 and other differentially expressed genes found that the CD47-related GO pathways were mainly pathways that regulate the process of actin cytoskeleton construction.The increased expression of genes related to these pathways including FAM49B,CLIP2,DIAPH1 and F2RL1 was related to the increased expression of CD47,and their increased expression was related to the poor prognosis of endometrial cancer;③The expression of CD47,FAM49B and CLIP2 was significantly increased in high-staging endometrial cancer.The above results indicate that CD47 and its related pathways that regulate actin cytoskeleton play an important role in the progression of endometrial cancer;CD47 may be a useful biomarker for prognosis and a potential immunotherapy target for endometrial cancer.
