摘　　要：目的探讨非小细胞肺癌(NSCLC)组织的环状RNA circ_0080220水平和临床意义。方法选取2015年1月至2019年12月本院手术切除的115对NSCLC患者癌组织和配对癌旁组织,采用实时荧光定量PCR(qPCR)检测NSCLC组织的circ_0080220水平并分析circ_0080220水平与患者临床病理特征和预后间的关系。结果NSCLC组织的circ_0080220水平为3.485±0.938,高于对应癌旁组织的1.043±0.460(t=25.068,P<0.001)。组织circ_0080220水平与NSCLC患者的年龄、性别、ECOG PS评分、病理类型和吸烟史均无关(P>0.05),而与TNM分期、肿瘤大小、淋巴结转移和分化程度有关(P<0.05)。NSCLC患者中circ_0080220高水平组的中位总生存期为44.0个月,短于低水平组的55.0个月(P=0.029),多因素分析显示,circ_0080220高水平是NSCLC预后不良的危险因素(HR=1.652,95%CI:1.123~2.247,P=0.035)。结论组织circ_0080220高水平与NSCLC患者的临床进展和预后较差密切相关,有作为NSCLC诊治和预后评估候选指标的潜能。Objective To investigate the level of circRNA circ_0080220 in non-small cell lung cancer(NSCLC)tissues and its clinical significance.Methods One hundred and fifteen pairs of cancer tissues and matched paracancerous tissues from NSCLC patients between January 2015 and December 2019 were collected.Real-time fluorescent quantitative PCR(qPCR)was used to detect circ_0080220 level in NSCLC tissues.Relationship of circ_0080220 level with clinicopathological characteristics and prognosis were analyzed.Results The circ_0080220 level was 3.485±0.938 in NSCLC tissues,higher than 1.043±0.460 in the corresponding adjacent tissues(t=25.068,P<0.001).Circ_0080220 level was not related to age,gender,ECOG PS,pathological type and smoking history(P>0.05),but related to TNM stage,tumor size,lymph node metastasis and differentiation(P<0.05).The median overall survival of circ_0080220 high-level group in patients with NSCLC was 44.0 months,shorter than 55.0 months of low-level group(P=0.029).High level of circ_0080220 was a risk factor for poor prognosis of NSCLC(HR=1.652,95%CI:1.123-2.247,P=0.035).Conclusion High tissue level of circ_0080220 is closely related to clinical progress and poor prognosis of NSCLC patients,which has the potential to be used as a candidate index for diagnosis,treatment and prognosis evaluation of NSCLC.
