摘 要:目的观察疏肝饮挥发油、黄酮、皂苷对肠易激综合征(irritable bowel syndrome,IBS)大鼠肠道动力的影响及对结肠组织中c-kit和干细胞生长因子(stem cell factor,SCF)表达的影响,明确疏肝饮治疗IBS的相关机制。方法36只Wistar雄性大鼠随机分为N组(正常)、M组(模型)、W组(水提物)、V组(挥发油)、F组(黄酮)、S组(皂苷),每组6只。除N组外,其余各组采用慢性避水应激法诱导IBS模型。在造模第3 d及药物干预第7 d观察各组大鼠在造模1 h内的排便粒数。从造模第4 d起,各组给予相应药物灌胃,连续给药7 d。观察各组大鼠的排便情况;末次给药后,评价各组大鼠肠道动力情况,分离结肠组织,采用免疫组化、Western blot法检测各组大鼠结肠c-kit和SCF蛋白表达。结果(1)V组和W组排便粒数与M组比较,明显减少(P<0.001);M组大鼠的玻璃球排出时间与N组相比,明显缩短(P<0.01);W组和V组玻璃球排出时间与M组相比,明显延长(P<0.01);而F组和S组玻璃球排出时间与M组比较,无显著差异(P>0.05)。(2)免疫组化结果表明M组大鼠结肠组织中c-kit和SCF蛋白表达比N组明显增多(P<0.05);W组和V组大鼠结肠组织c-kit和SCF的蛋白表达相较M组的表达显著下降(P<0.05),而F组和S组c-kit和SCF的蛋白表达与M组相比无明显变化(P>0.05)。Western blot结果表明M组大鼠结肠组织中c-kit和SCF蛋白表达比N组明显增多(P<0.001,P<0.01);与M组比较,W组和V组大鼠结肠组织c-kit的蛋白表达显著下降(P<0.001,P<0.01);W组和V组的SCF蛋白表达相较M组的表达亦显著下降(P<0.05),而F组和S组c-kit和SCF的蛋白表达与M组相比无明显变化(P>0.05)。结论疏肝饮挥发油和水提物可减缓腹泻型IBS大鼠肠道动力异常,其机制可能与下调结肠组织中c-kit和SCF蛋白的表达有关。Objective To observe the effects of effective components of Shugan Decoction on intestinal motility and expression of c-kit and SCF in colon of irritable bowel syndrome(IBS) model rats. Methods Thirty-six male wistar rats were randomly divided into normal group(group N), model group(group M), Shugan Decoction water extract group(group W), Shugan Decoction volatile oil group(group V), Shugan Decoction flavonoid group(group F), Shugan Decoction saponin group(group S), with six rats in each group. In addition to the normal group, water avoidance stress(WAS) was used to induce IBS model. From the 4 th day, each drug group was given the corresponding extract intervention, and the group N and group M were given the same volume of 0.9%NaCl solution, and the drug was continuously administered by intragastric administration for 7 d. After the last administration, the intestinal function of each group was evaluated by measuring the excretion time of the vitreous glomerulus.The expression of c-kit and SCF in colon of rats in each group were detected by immunohistochemistry and Western blot. Results(1)After drug intervention, compared with the group M, the number of defecation granules in group V and group W decreased significantly(P<0.001);The time of vitreous bulb excretion in group M was significantly shorter than that in group N(P<0.01);Compared with group M, the time of glass ball excretion in group W and group V was significantly longer(P<0.01);However, there was no significant difference in the time of glass ball discharge between group F and group S(P>0.05).(2)The results of immunohistochemistry showed that the expression of c-kit and SCF protein in colon tissue of group M was significantly higher than that of group N(P<0.05);The protein expressions of c-kit and SCF in colonic tissues of group W and group V were significantly lower than those of group M(P<0.05), while the protein expressions of c-kit and SCF in group F and group S were not significantly different from those of group M(P>0.05).
