摘　　要：目的研究姜黄素对人肺癌细胞株A549迁移能力的影响,并分析SRPK1的可能作用机制。方法采用细胞迁移实验检测姜黄素对人肺癌A549细胞迁移能力的影响,并采用Western blot法和RT-PCR检测SRPK1的表达。siRNA法建立SRPK1表达减低的A549+siRNA-SRPK1细胞系,细胞迁移实验比较A549+siRNA-SRPK1细胞与A549细胞的迁移能力,并采用Western blot法和RT-PCR检测细胞中SRPK1、ZEB1、TWIST1、TGF-β和SNAIL等肿瘤转移相关基因的蛋白和mRNA表达情况。结果姜黄素能显著抑制人肺癌A549细胞的迁移;姜黄素能显著下调SRPK1的表达水平;siRNA-SRPK1处理后,A549细胞迁移能力显著下降,加入姜黄素后迁移能力下降更明显。结论姜黄素能通过下调SRPK1的表达来抑制肺癌A549细胞的迁移。Objective To study the effect of Curcumin on migration ability of human lung cancer cell line A549 and to analyze the potential mechanism of SRPK1.Methods The effect of Curcumin on migration ability of human lung cancer A549 cells was detected by cell migration examination,and expression of SRPK1 gene was detected by Western blot and real time-polymerase chain reaction(RT-PCR)methods.The A549+siRNA-SRPK1 cell line with reducing SRPK1 expression was established by the siRNA method.Migration abilities of A549+sirna-SRPK1 cell and A549 cells were compared by using cell migration examination.Protein and mRNA expressions of tumor metastasis related genes such as SRPK1,ZEB1,TWIST1,TGF-βand SNAIL were detected by Western blot and RT-PCR methods.Results Curcumin significantly inhibited the migration of human lung cancer A549 cell and significantly down-regulated the expression of SRPK1.After treatment by siRNA-SRPK1,the migration ability of A549 cells was significantly decreased,and the decrease was more obvious after treatment by Curcumin.Conclusion Curcumin may inhibit the migration of lung cancer cell line A549 by down-regulating the expression of SRPK1.
