摘 要:目的探讨黄连素通过调控Hippo信号通路抑制人胃癌细胞系HGC-27的作用机制。方法用5、10μmol/L浓度黄连素处理人胃癌细胞系HGC-2724 h后,利用Real-time PCR和Western blot检测Hippo信号通路重要分子表达水平的变化。结果在mRNA水平上,与对照组相比,黄连素组TEA domain transcription factors 3(TEAD3)与TEAD4表达水平显著下降(5μmol/L组P<0.05;10μmol/L组P<0.01),呈浓度梯度依赖性下降趋势;黄连素10μmol/L组Yes-associated protein(YAP)与CyclinE1表达水平显著低于对照组(P<0.01),而MST1表达水平显著高于对照组(P<0.05)。在蛋白水平上,与对照组相比,黄连素10μmol/L组TEAD4、YAP与CyclinE1表达水平显著下降(P<0.05,或P<0.01);MST1与p-YAP表达水平显著上升(P<0.05)。结论黄连素可能通过调控Hippo信号通路抑制胃癌细胞HGC-27,发挥抗肿瘤作用。Objective To study the effect of berberine in inhibition of human gastric cancer HGC-27 cells by regulating Hippo signaling pathway.Methods After 5 and 10μmol/L berberine treatment of HGC-27 cells for 24 h,real-time PCR and western blot were used to detect the expression of important molecules of Hippo signaling pathway.Results Compared with the control group,the levels of TEAD3 and TEAD4 mRNA were significantly decreased in two Berberine groups(5μmol/L group,P<0.05;10μmol/L group,P<0.01).The expression of YAP and CyclinE1 was significantly decreased in 10μmol/L Berberine group(P<0.01),while the expression of MST1 was higher than that in the control group(P<0.05).At the protein level,the expression of TEAD4,YAP and CyclinE1 were significantly decreased in 10μmol/L Berberine group(P<0.05,P<0.01),while the expression of MST1 and p-YAP were significantly higher than that in the control group(P<0.05).Conclusion Berberine may inhibit gastric cancer HGC-27 cells by regulating Hippo pathway.
