摘 要:目的观察过量维生素D_(3)(VitD_(3))灌胃的Wistar大鼠毒性反应及组织器官钙化情况。方法将6~8周龄Wistar雄性大鼠分为给药组和对照组,每组20只,给药组给予VitD_(3)70万U/kg灌胃,对照组同法给予等体积蒸馏水,隔天1次,共灌胃2次。灌胃后记录大鼠一般情况,并于灌胃结束1周后取大鼠心脏、肝脏、肾脏、肺、血管进行HE染色,观察组织病理变化;茜素红染色观察组织器官钙化情况;qRT-PCR法检测骨形态发生蛋白2(BMP2)mRNA,免疫组化法检测BMP2蛋白。结果给药组大鼠精神萎靡,进食量减少,毛发无光泽,口角有血迹,出现咖啡色尿,解剖后发现弥漫性肺出血及肠梗阻现象。HE染色及茜素红染色观察到给药组大鼠心脏、肾脏、肺、血管均出现不同程度的钙化灶等病理改变,而肝脏中未观察到病理改变。给药组心脏、肾脏、肺、血管中BMP2 mRNA及蛋白表达水平均高于对照组(P均<0.05)。结论过量VitD_(3)灌胃可引起Wistar大鼠急性毒性反应及多种组织器官钙化。Objective To observe the toxicity and organ calcification in Wistar rats with vitamin D_(3)(VitD_(3))over⁃dose.Methods Forty 6-8 weeks old male Wistar rats were divided into two groups:the VitD_(3) treated group and the nor⁃mal control group,with 20 rats in each group.The rats in the VitD_(3) treated group were given VitD_(3)7×105 U/kg by gavage,while the rats in the normal control group were given the same volume of distilled water,once every other day,twice in to⁃tal.The general condition of rats was recorded after gavage.The heart,liver,kidney,lung and blood vessels were exam⁃ined by HE staining to observe the histopathological changes;the calcification of tissues was observed by alizarin red stain⁃ing;the mRNA and protein expression levels of bone morphogenetic protein 2(BMP2)were detected by qRT-PCR and im⁃munohistochemistry.Results After administration with VitD_(3),the rats became depressed,with less food intake,dull hair and brown urine.Blood stains occurred at the corners of their mouth.After dissection,the diffuse pulmo⁃nary hemorrhage and intestinal obstruction were found.The results of HE staining and alizarin red staining showed calcifi⁃cation in the heart,kidney,lung and blood vessels,but no changes in liver.Results of qRT-PCR and immunohistochemis⁃try showed that:compared with the normal control group,the mRNA and protein expression levels of BMP2 in the heart,kidney,lung and blood vessels were higher in the VitD_(3) treated group(all P<0.05).Conclusion Overdose VitD_(3) could cause acute toxic symptoms and organ calcification in Wistar rats.
