摘　　要：目的探讨食管鳞状细胞癌(esophageal squamous cell carcinoma,ESCC)组织中长链非编码RNA(LncRNA)linc00342的表达及临床意义。方法选取2015年6月至2016年9月于赣榆区人民医院接受ESCC根治术的89例ESCC患者的癌组织及癌旁组织为研究对象,采用实时荧光定量PCR检测LncRNA linc00342的表达水平,并分析肿瘤组织中LncRNA linc00342的表达水平与患者临床病理参数的关系;ROC曲线评价LncRNA linc00342对ESCC的诊断价值;采用Kaplan-Meier法进行生存分析;多因素Cox回归分析影响ESCC患者预后不良的危险因素。结果ESCC患者癌组织和癌旁组织中LncRNA linc00342的相对表达量分别为3.16±1.27和1.64±0.51,二者差异有统计学意义(t=10.478,P<0.001);LncRNA linc00342的表达水平与TNM分期(χ^(2)=2.592,P=0.011)、淋巴结转移(χ^(2)=2.240,P=0.028)有关;Kaplan-Meier生存分析显示,LncRNA linc00342高表达和低表达患者的累积生存率分别为21.74%、55.81%,差异有统计学意义(Log-rankχ^(2)=20.896,P=0.000);单因素分析显示,TNM分期(χ^(2)=8.616,P=0.003)、淋巴结转移(χ^(2)=4.732,P=0.030)及LncRNA linc00342表达(χ^(2)=17.092,P=0.000)是影响ESCC患者预后不良的危险因素;LncRNA linc00342表达预测ESCC的ROC曲线下面积为0.903(95%CI:0.874~0.938,P<0.001);多因素Cox回归分析结果显示,临床分期为Ⅲ~Ⅳ期(HR=1.943,95%CI:1.280~2.950,P=0.002)、淋巴结转移(HR=1.884,95%CI:1.241~2.860,P=0.003)、LncRNA linc00342高表达(HR=2.107,95%CI:1.361~3.262,P=0.001)是影响ESCC患者预后不良的独立危险因素。结论LncRNA linc00342在ESCC组织中表达上调,是影响ESCC发生及预后不良的潜在生物学指标。Objective To investigate the expression and clinical significance of long non-coding RNA(LncRNA)linc00342 in esophageal squamous cell carcinoma(ESCC).Methods The tumor tissue and adjacent normal tissue samples from 89 ESCC patients admitted in our hospital for radical operation during June 2015 and September 2016 were collected,and the expression levels of LncRNA linc00342 in these samples were detected by the real-time fluorescence quantitative PCR.The relationship between the expression level of LncRNA linc00342 in ESCC tissues and clinicopathological parameters was analyzed.The diagnostic value of LncRNA linc00342 for ESCC was evaluated by the receiver operating characteristic curve(ROC).The survival time and risk factors influencing prognosis of ESCC patients were analyzed by the Kaplan-Meier method and multivariate Cox regression analysis,respectively.Results The relative expression levels of LncRNA linc00342 in the tumor tissues and adjacent normal tissues of ESCC patients were 3.16±1.27 and 1.64±0.51,respectively,and there was significant difference between them(t=10.478,P<0.001).The expression levels of LncRNA linc00342 were correlated with TNM stage(χ^(2)=2.592,P=0.011)and lymph node metastasis(χ^(2)=2.240,P=0.028).The Kaplan-Meier survival analysis showed that the cumulative survival rates of the patients with high and low expression of LncRNA linc00342 were 21.74%and 55.81%,respectively,and there was significant difference between them(log-rankχ^(2)=20.896,P=0.000).Theunivariate analysis showed that TNM stage(χ^(2)=8.616,P=0.003),lymph node metastasis(χ^(2)=4.732,P=0.030)and the expression of LncRNA linc00342(χ^(2)=17.092,P=0.000)were the risk factors influencing the prognosis of ESCC patients.The area under ROC curve of the expression level of LncRNA linc00342 in the prediction of ESCC was 0.903(95%CI:0.874-0.938,P<0.001).The multivariate Cox regression analysis showed that the clinical stageⅢ-Ⅳ(HR=1.943,95%CI:1.280-2.950,P=0.002),lymph node metastasis(HR=1.884,95%CI:1.241-2.860,P=0.003)an
