摘  要:[目的]本文基于茄亲本及F2材料高通量测序数据,开发茄果皮着色光敏性相关dCAPS标记,为标记辅助选择及果皮着色光敏性基因定位提供理论依据。[方法]以果皮着色光敏感茄品种‘蓝山禾线’和光不敏感茄品种‘145’及其60个F2单株为材料,通过高通量简化基因组测序技术和群体分离分析法(BSA)相结合,在候选区间内设计dCAPS引物,开发多态性共显性标记,并使用192个F2单株和123个茄品种对标记进行验证。[结果]茄果皮着色光敏性为单基因控制的显性遗传。根据BSA-seq和关联分析,当置信度为0.99时,在第10号染色体上鉴定出3个候选区段87.080~88.090 Mb、91.475~92.475 Mb、92.785~93.785 Mb,总长度为3.01 Mb。在候选区段开发了1个可用于区分果皮着色光敏感和光不敏感茄的dCAPS-9标记,该标记在192个F2单株和123个茄品种中的表型对应度分别达到98.4%和91.9%。[结论]dCAPS-9在分离群体及品种鉴定中具有较高的准确性,对于茄果皮着色光敏性标记辅助选择具有重要意义。[Objectives]The aim of this study was to develop dCAPS molecular markers for photosensitivity of anthocyanin production in eggplant(Solanum melongena L.)based on high-throughput re-sequencing data.The molecular marker can be used for molecular assisted selection and gene mapping analysis.[Methods]In this study,photosensitive eggplant variety‘Lanshanhexian’,non-photosensitive eggplant variety‘145’and 60 F2 population were used as materials.With reduced-representation sequencing and bulked segregant analysis(BSA),polymorphic dCAPS primers were designed.A further testing of markers was performed between 192 F2 populations and 123 eggplant accessions to verify the accuracy.[Results]The photosensitivity of eggplant fruit color was dominant inheritance controlled by a single gene,and three candidate segments with a total length of 3.01 Mb were obtained by BSA-seq and correlation analysis:87.080-88.090 Mb,91.475-92.475 Mb,92.785-93.785 Mb.A molecular marker dCAPS-9 was developed,which could be used to distinguish photosensitive and non-photosensitive eggplants.The further testing showed the selection accuracy of 98.4%and 91.9%between marker test and phenotype in 192 F2 and 123 varieties of eggplant,respectively.[Conclusions]dCAPS-9 had high accuracy both in the F2 populations and eggplant varieties.The development of dCAPS-9 was of great significance for the photosensitive molecular marker-assisted breeding in eggplant.
