摘 要:本实验旨在研究不同浓度没食子酸(GA)对猪精液常温(17℃)保存效果的影响并筛选没食子酸的浓度范围。在猪精液稀释剂中分别添加不同浓度(0、5、6、7、8、9、10μg/mL)没食子酸,于不同保存时间测定猪精子的活率、AB级精子数、质膜完整率及顶体完整率指标。结果表明,保存第1~3 d各浓度组间指标无明显差异(P>0.05),保存第5 d,6、9μg/mL组除组质膜完整率极显著高于对照组(P<0.01),保存第5、7 d时,7、9μg/mL组精子活率显著高于对照组(P<0.05);保存至第9 d时9μg/mL组活率显著高于对照组(P<0.05)。综上表明,一定浓度范围的没食子酸有助于提高常温(17℃)保存下猪精液的质量,并延长保存时间。The present study aimed to investigate the effects of different concentrations of gallic acid(GA)on the preservation of boar sperm at 17℃,and the optimum addition concentration was screened out.Different concentrations(0,5,6,7,8,9,10μg/mL)of GA were added into Beltsville thawing solution.During the process of liquid preservation at 17°C the sperm motility,grade A+B sperm number,plasma membrane integrity and acrosome integrity were measured at different times.The results showed that there was no significant difference in sperm viability,plasma membrane integrity and acrosome integrity between the groups at the 1st and 3rd day of preservation(P>0.05).The sperm viability of the 6 and 9μg/mL groups was significantly higher than that of the control group,when stored for 5th day(P<0.01).The sperm viability of the 7 and 9μg/mL groups was significantly higher than that of the control group,when stored for the 5th and 7th day(P<0.05).The 9μg/mL activity was significantly higher than that of the control group,when stored for 9th day(P<0.05).In conclusion,it is shown that GA in a certain concentration range can help improve the quality of pig semen stored at room temperature(17°C)and extend the storage time.
