摘 要:目的:检测钙网蛋白(CRT)、驱动蛋白超家族蛋白5A(KIF5A)在三阴性乳腺癌组织中的表达,并分析CRT、KIF5A蛋白表达与临床病理特征及预后的关系。方法:收集2013年1月—2014年12月125例三阴乳腺癌患者的癌组织和癌旁正常组织的石蜡样本,患者均为女性。采用免疫组织化学法检测CRT、KIF5A蛋白的表达情况,分析癌组织中CRT、KIF5A蛋白表达与患者临床病理特征之间的关系,Kaplan Meier法分析CRT、KIF5A蛋白表达对患者术后5年总生存率的影响。结果:三阴性乳腺癌组织中,CRT、KIF5A蛋白的阳性表达率为54.40%(68/125)、60.00%(75/125),均分别高于癌旁正常组织的8.80%(11/125)、10.40%(13/125),差异有统计学意义(P<0.05)。三阴性乳腺癌组织中,CRT蛋白表达与肿瘤大小、淋巴结转移和TNM分期相关(P<0.05);KIF5A蛋白表达与肿瘤大小、淋巴结转移、远处转移和TNM分期相关(P<0.05)。Kaplan Meier结果显示,CRT阳性表达组的术后5年总生存率低于CRT阴性表达组(P<0.05),KIF5A阳性表达组的术后5年总生存率低于KIF5A阴性表达组(P<0.05)。结论:三阴性乳腺癌组织中CRT和KIF5A蛋白阳性表达率高,且均与患者不良临床病理特征和较低术后5年总生存率相关。CRT和KIF5A具有作为三阴乳腺癌治疗靶标的潜能。Objective: To detect the protein expressions of calreticulin(CRT) and kinesin super family protein 5 A(KIF5 A) in the triple negative breast cancer tissues, and to investigate their correlations with the clinicopathological characteristics and the prognosis. Methods: The cancer and adjacent paraffin tissues from the 125 cases of patients with triple negative breast cancer were collected from January 2013 to December 2014, all patients were female. The protein expressions of CRT and KIF5 A in the paraffin tissues were evaluated by the immunohistochemistry assay, the correlations between the CRT/KIF5 A protein expression in cancer tissues and the clinicopathological characteristics were analyzed, the Kaplan Meier was utilized to evaluate the effects of CRT/KIF5 A protein expression on the post-surgery 5-year overall survival rate. Results: In the triple negative breast cancer tissues, the positive rates of CRT and KIF5 A protein were 54.40%(68/125), 60.00%(75/125),which were higher than 8.80%(11/125), 10.40%(13/125) in the adjacent normal tissues, respectively(P<0.05). In the triple negative breast cancer tissues, the CRT protein expression was correlated with tumor size, lymph node metastasis and the TNM stages(P<0.05). Besides, the KIF5 A protein expression was correlated with tumor size, lymph node metastasis, distance metastasis and the TNM stages(P<0.05). Kaplan Meier results suggested that the post-surgery 5-year overall survival rate of the CRT-positive group was much lower than that in the CRT-negative group(P<0.05), while the post-surgery 5-year overall survival rate of the KIF5 A-positive group was much lower than that in the KIF5 A-negative group(P<0.05). Conclusions: The positive rates of CRT and KIF5 A protein in the breast cancer tissues were upregulated, which were both correlated with the poor clinicopathological characteristics and low 5-year overall survival rate. CRT and KIF5 A may serve as the therapeutic biomarkers for the breast cancer.
