摘  要:目的研究定志小丸有效成分中人参皂苷Rb3联合β-细辛醚对血管性痴呆模型小鼠的作用。方法采用四血管阻断法建立血管性痴呆小鼠模型后,随机分为模型组、人参皂苷Rb3组(10 mg/kg)、β-细辛醚组(10 mg/kg)、人参皂苷Rb3联合β-细辛醚组(10 mg/kg+10 mg/kg)、盐酸多奈哌齐组(1 mg/kg)和LY294002组(1 mg/kg),每组10只。除假手术组(n=10)和模型组给予等量体积生理盐水,各组给药30 d,每天2次。采用ELISA法检测各组小鼠海马和血清中3-硝基酪氨酸(3-NT)、神经营养因子-3(NT-3)、聚腺苷酸二磷酸核糖基聚合酶1(PARP1)和醛糖还原酶(AR)的含量,采用HE法观察海马部位组织病理学的变化。结果与假手术组比较,模型组的海马和血清中3-NT、NT-3、PARP1和AR含量显著增加(P<0.01);与模型组比较,人参皂苷Rb3组、β-细辛醚组、人参皂苷Rb3联合β-细辛醚组和盐酸多奈哌齐组的海马和血清中3-NT、NT-3、PARP1和AR含量显著减少(P<0.05),而LY294002组的海马和血清中3-NT、NT-3、PARP1和AR含量显著增加(P<0.01);与人参皂苷Rb3组和β-细辛醚组比较,人参皂苷Rb3联合β-细辛醚组的海马和血清中3-NT、NT-3、PARP1和AR含量显著增加(P<0.05),而盐酸多奈哌齐组的海马和血清中3-NT、NT-3、PARP1和AR含量显著减少(P<0.05)。除LY294002组,其他各给药组的海马组织病理损伤均有所减轻。结论定志小丸有效成分中人参皂苷Rb3联合β-细辛醚具有一定神经保护作用,表现在可以减少血管性痴呆模型小鼠海马和血清中3-NT、NT-3、PARP1和AR含量,以及减缓海马组织损伤,而联合用药的作用优于单独给药组。Objective To study the effect of ginsenoside Rb3 andβ-asarone in active ingredients of Dingzhi Xiaowan Decoction(定志小丸)on vascular dementia model mice.Methods After establishing a mouse model of vascular dementia using the four-vessel blocking method,they were randomly divided into model groups,ginsenoside Rb3(10 mg/kg),β-asarone group(10 mg/kg),ginsenoside Rb3 combined withβ-asarone the combination administration of ginsenoside Rb3 andβ-asarone group(10 mg/kg+10 mg/kg),the donepezil hydrochloride group(1 mg/kg)and the LY294002 group(1 mg/kg),10 rats in each group.Except for the sham operation group(n=10)and the model group,an equal volume of normal saline was administered,and each group was administered for 30 days,twice a day.Detection of 3-nitrotyrosine(3-NT),neurotrophic factor-3(NT-3),polyadenylic acid diphosphate ribose polymerase 1(PARP1)and aldose reductase(AR)content in hippocampus and serum of each group of mice by ELISA,the histopathological changes of hippocampus were observed by HE method.Results Compared with the sham operation group,the levels of 3-NT,NT-3,PARP1 and AR in the hippocampus and serum of the model group increased significantly(P<0.01);compared with the model group,the content of 3-NT,NT-3,PARP1 and AR in hippocampus and serum of the ginsenoside Rb3 group,theβ-asarone group and combination administration of ginsenoside Rb3 andβ-asarone group and the donepezil hydrochloride group were significantly reduced(P<0.05),while the contents of 3-NT,NT-3,PARP1 and AR were significantly increased in LY294002 group(P<0.01);compared with the ginsenoside Rb3 group andβ-asarone group,3-NT,NT-3,PARP1 and AR in the hippocampus and serum of the combination administration of ginsenoside Rb3 andβ-asarone group were significantly increased(P<0.05),while the contents of 3-NT,NT-3,PARP1 and AR in the hippocampus and serum of the donepezil hydrochloride group were significantly reduced(P<0.05).Excepted for the LY294002 group,all other histopathological damage in the hippocampus of the administra
