摘 要:云南火焰兰(Renanthera imschootiana Rolfe)是濒临灭绝的附生植物,关于其分子水平的研究少有报道。为了获得云南火焰兰相关基因组基础信息,本研究通过Illumina HiSeqTM4000技术对云南火焰兰叶片进行转录组测序,并利用相关生物信息学数据库对其进行序列比对和数据分析。分析结果表明:共获得77888条功能基因(Unigenes),其N50为1348 bp,平均长度为805 bp,Q20和Q30序列分别占98.04%和94.32%。经比对,在Nr、KEGG、KOG、Swiss-prot、GO等数据库中能得到注释的Unigene分别为31298、29356、14852、15697、17645条。在GO数据库中,共得到24473个GO功能,将其分为3个大类和47个亚类,以生物过程这一类别所注释的基因数最多;KOG功能注释信息有17743个,将其分为25个基因功能大类,其中基因数目较多的分别是一般功能基因和信号传导机制的相关基因;在KEGG中涉及的相关通路可将其归为19个亚类,其中以代谢通路较为突出。此外,对云南火焰兰转录组Unigene进行CDS比对和预测,共预测到2317条CDS,其大小主要集中在200-500 bp,占总CDS的79.41%。本研究可为云南火焰兰乃至火焰兰属植物功能基因的挖掘、利用以及遗传育种等方面的工作提供一定的科学依据。Renanthera imschootiana Rolfe is an endangered epiphyte,and there are few reported on its molecular biology.In order to obtain the basic genomic information related to R.imschootiana Rolfe,Illumina HiSeqTM4000 technology was used in this study to conduct transcriptome sequencing of R.imschootiana Rolfe leaves,then sequence comparison and data analysis were carried out using related bioinformatics database.The results showed that 77888 Unigenes were obtained,with N50 of 1348 bp.The average length was 805 bp,Q20 and Q30 sequences accounted for 98.04%and 94.32%respectively.By comparison,31298,29356,14852,15697 and 17645 Unigenes were annotated in database of Nr,KEGG,KOG,Swiss-prot and GO,respectively.In the GO database,a total of24473 functions were obtained,and were divided into 3 major categories and 47 subcategories,in which the biological processes were the category with the most annotated genes.In the KOG database,17743 functional annotation information were found,and were divided into 25 gene function categories.Among them,the genes with a high number of genes were the general functional genes and the genes about signal transduction mechanism.The related pathways involved in KEGG could be classified into 19 subclasses,among which metabolic pathways were abundant.In addition,the ESTS can software was used to perform CDS comparison and prediction about the transcriptome Unigenes of R.imschootiana Rolfe,and a total of 2317 CDS were predicted,the size of which was mainly distributed between 200 and 500 bp,accounting for 79.41%of the total CDS.Theoretical basis could be provided for the work of mining,utilization of functional genes and genetic breeding on plant of R.imschootiana Rolfe even renanthera genus.
