摘 要:为研究木霉粗酶液胞外分泌物的生物活性,从愈创木酚的用量、反应温度、缓冲液种类、缓冲液pH、缓冲液浓度5个因素,对木霉粗酶液胞外分泌物漆酶的酶活力进行研究;从反应时间、反应温度、缓冲液pH、染料浓度、缓冲液浓度、酶液稀释倍数6个因素对木霉粗酶液的胞外分泌物漆酶对染料的脱色作用进行研究。结果表明:木霉漆酶在反应温度为25℃,琥珀酸缓冲液的pH为4.0,琥珀酸缓冲液浓度为0.045 mol/L,愈创木酚用量为0.04 mol/L时,木霉粗酶液胞外分泌物漆酶酶活力最优;木霉粗酶液的胞外分泌物漆酶其脱色作用在反应温度为30℃,pH为3.5,染料浓度为0.01 g/L,醋酸缓冲液浓度为0.2 mol/L时,木霉粗酶液的胞外分泌物漆酶脱色作用最好。In order to study the biological activity of extracellular secretion of Trichoderma crude enzyme solution, enzyme activity of laccase from the extracellular secretion of Trichoderma crude enzyme was studied from five factors including dosage of guaiacol, reaction temperature, type of buffer solution, pH of buffer solution and concentration of buffer solution. Meanwhile, decolorization effect of laccase from extracellular secretion of Trichoderma crude enzyme solution was studied from six factors including reaction time, reaction temperature, buffer pH, dye concentration, buffer concentration and dilution ratio of enzyme solution. The results showed that when reaction temperature was 25 ℃, pH value of succinic acid buffer was 4.0, concentration of succinic acid buffer was 0.045 mol/L, and dosage of guaiacol was 0.04 mol/L, laccase activity of extracellular secretion of Trichoderma crude enzyme was the best;when reaction temperature was 30 ℃, pH value was 3.5,concentration of dye was 0.01 g/L, and concentration of acetic acid buffer was 0.2 mol/L, decolorization effect of laccase from extracellular secretion of Trichoderma crude enzyme was the best.
