摘　　要：目的:研究长链非编码RNA H19(LncRNA H19,H19)及缺氧诱导因子-1α(HIF-1α)对口腔鳞癌细胞增殖侵袭及细胞周期的影响。方法:纳入口腔鳞癌(OSCC)患者45例,分析肿瘤及癌旁组织中H19及HIF-1α的表达差异及相关性,同时建立上调及干扰OSCC细胞Cal-27中H19的表达,观察细胞增殖、侵袭及细胞周期的变化。结果:H19在OSCC组织中高表达,并与HIF-1α表达正相关,H19可促进OSCC细胞增殖、侵袭及细胞周期进程,上调Cal-27细胞中H19的表达显著上调;HIF-1α干扰Cal-27细胞中H19表达降低了细胞中HIF-1α的表达。结论:LncRNA H19通过上调HIF-1α的表达促进人口腔鳞癌细胞增殖侵袭能力及细胞周期。Objective:To study the effects of long non-coding RNA H19(LncRNA H19,H19)and Hypoxia inducible factor-1α(HIF-1α)on the proliferation,invasion and cell cycle progression of human oral squamous cell carcinoma(OSCC)cells.Methods:45 patients with OSCC were included.The difference and correlation between the expression H19 and HIF-1αin tumor and adjacent tissues,in the OSCC cell lines of SCC-4,SCC-9,SCC-25,CAL-27,TCA8113 and KB were examined.The expression of H19 in Cal-27 cells was up-regulated and interfered by RNA interference(RNAi)for the obseration of cell proliferation,invasion and cell cycle distribution changes.Results:H19 was highly expressed in OSCC tissues and OSSC cell lines,and positively correlated with HIF-1αexpression.In vitro,H19 could promote malignant proliferation,invasive ability and cell cycle progression of OSCC cells.Upregulation of H19 in Cal-27 cells significantly up-regulated HIF-1αexpression in the cells and down regulation of H19 decreased HIF-1αin the cells.Conclusion:H19 promotes proliferation,invasion and cell cycle progression of human OSCC cells via up-regulating expression of HIF-1α.
