摘  要:目的:探讨RNAi抑制HIF-1α对口腔鳞癌生长和血管生成的影响。方法:利用RT-PCR、Western blot和IHC检测12例OSCC和正常组织临床标本中HIF-1α的表达;建立30只裸鼠OSCC动物模型,利用RNAi抑制HIF-1α的表达,观察肿瘤体积生长的变化并检测肿瘤凋亡细胞;检测肿瘤组织中VEGF的表达和微血管密度(MVD)的变化。结果:OSCC临床标本中HIF-1α的表达明显高于正常组织(P<0.05);在HIF-1α被抑制之后,荷瘤裸鼠肿瘤的生长速度明显低于对照组(P<0.05),且实验组肿瘤组织的凋亡细胞明显增加(P<0.05),VEGF的表达和MVD明显降低(P<0.05)。结论:HIF-1α在OSCC肿瘤生长和血管生成中有促进作用。Objective:To investigate the effects of HIF-1αRNAi on the growth and angiogenesis of oral squamous cell carcinoma.Methods:The expression of HIF-1αin 12 clinical specimens of OSCC and normal tissue was detected by RT-PCR,Western blot and IHC.The animal model of OSCC was established in 3D nude mice,the expression of HIF-1αwas inhibited by RNAi,the change of tumor volume growth was observed and the apoptotic cells were detected,and the expression of VEGF and the change of microvessel density(MVD)in tumor tissue were detected.Results:The expression of HIF-1αin OSCC clinical specimens was significantly higher than that in normal tissue(P<0.05),after HIF-1αRNAi,the growth rate of tumor in nude mice was significantly lower than that in the controls(P<0.05),and the apoptotic cells in tumor tissue in the experimental group increased(P<0.05),while the expression of VEGF and MVD decreased(P<0.05).Conclusion:HIF-1αmay play a role in tumor growth and angiogenesis of OSCC.
