骨保护素(OPG)基因多态性与种植体周围组织疾病的相关性研究

摘　　要：目的:探讨骨保护素(OPG)基因A163G位点(rs3102735)单核苷酸多态性与种植体周围组织疾病的相关性。方法:纳入种植体周围组织疾病患者和健康种植体患者各80例,在颊黏膜脱落的上皮细胞中提取DNA后,采用PCR-Sanger法检测患者OPG-A163G位点,分析其与种植体周围组织疾病发病风险的关系。结果:OPG-A163G位点的基因型GG、GA、AA及等位基因G、A在健康种植体组和种植体周围组织疾病组中分布频率的差异有统计学意义(基因型:χ^(2)=12.33,P=0.00;等位基因:χ^(2)=14.05,P=0.00)。进一步分析,3种基因型及等位基因在健康种植体组和种植体周围炎组中分布频率的差异也有统计学意义(基因型:χ^(2)=16.10,P=0.00;等位基因:χ^(2)=19.16,P=0.00);在健康种植体组与种植体周围黏膜炎组的差异无统计学意义(P>0.05)。结论:OPG-A163G基因位点的多态性可能与种植体周围组织疾病的发病相关,G等位基因可能是发病的危险因素。Objective:To investigate the association between A163G(rs3102735)single nucleotide polymorphisms of osteoprotegerin(OPG)gene and the susceptibility of peri-implant diseases.Methods:Buccal mucosa swabs were scraped from 80 patients with peri-implant diseases and 80 subjects with healthy implants.PCR-Sanger sequencing was used to analyze the difference of the promoter region of OPG-A163G gene between the 2 groups,and the correlation between polymorphism and peri-implant disease was analyzed.Results:A statistically significant difference was found in the distribution frequencies of genotypes GG,GA,AA and alleles G,A of OPG-A163G between the 2 groups(genotypes:χ^(2)=12.33,P=0.00;alleles:χ^(2)=14.05,P=0.00).The 3 genotypes and the alleles were also significantly different between the 2 groups(genotypes:χ^(2)=16.10,P=0.00;alleles:χ^(2)=19.16,P=0.00).However,there was no significant difference between healthy implant group and peri-implant mucositis group(P>0.05).Conclusion:There is association between OPG-A163G single nucleotide polymorphisms and peri-implant disease and the allele G may be a risk factor for the disease.
