摘 要:目的探讨非小细胞肺癌(NSCLC)中星形胶质细胞上调基因-1(AEG-1)和E-钙黏素(E-cadherin)的表达及临床意义。方法选取2018年1月至2020年12月在湖州市中心医院行手术切除和石蜡包埋的NSCLC标本83例,并选取距肿瘤边缘5 cm以上的配对远端正常组织83例作为对照。采用免疫组化En Vision二步法检测NSCLC组织和远端正常组织中AEG-1和E-cadherin蛋白表达情况,分析NSCLC不同临床病理特征与AEG-1和E-cadherin表达的关系及两者蛋白表达的相关性。结果NSCLC组织中AEG-1的阳性表达率为63.86%,明显高于远端正常组织的9.64%,差异有统计学意义(P<0.01)。NSCLC组织中E-cadherin的阳性表达率为33.73%,明显低于远端正常组织的75.90%,差异有统计学意义(P<0.01)。NSCLC组织中AEG-1和E-cadherin阳性表达与患者的肿瘤TNM分期、分化程度、淋巴结转移均有关(均P<0.05)。NSCLC组织中AEG-1和E-cadherin蛋白表达呈负相关(r=-0.530,P<0.01)。结论NSCLC组织中AEG-1和E-cadherin蛋白表达呈负相关,两者负向调节共同促进了NSCLC的发生、发展,为NSCLC的治疗提供新的潜在靶点。Objective To investigate the expression of astrocyte elevated gene-1(AEG-1)and E-cadherin in non-small cell lung cancer(NSCLC)and their clinicopathological significance.Methods Cancer tissue and paired-adjacent normal tissue samples were obtained from 83 patients with NSCLC who underwent surgical resection at Huzhou Central Hospital from January 2018 to December 2020.The expression levels of AEG-1 and E-cadherin were detected by immunohistochemical method and the correlation between expression level and clinicopathological characteristics was analyzed.Results The positive expression rate of AEG-1 in NSCLC cancer tissue was significantly higher than that in adjacent normal tissue(63.86%vs.9.64%,P<0.01).The positive expression rate of E-cadherin protein in NSCLC cancer tissue was significantly lower than that in adjacent normal tissue(33.73%vs.75.90%,P<0.01).The positive expression of AEG-1 and E-cadherin was closely associated with tumor TNM stage,differentiation degree and lymph node metastasis in NSCLC(all P<0.05).The AEG-1 expression in NSCLC tissue was negatively correlated with E-Cadherin expression in NSCLC(r=-0.530,P<0.01).Conclusion The expression of AEG-1 is negatively correlated with E-cadherin in NSCLC,which may promote the occurrence and development of NSCLC and provide a potential target for the treatment of NSCLC.
