摘  要:目的探究上转换荧光纳米材料(upconversion nanoparticles,UCNP)BaGdF_(5)@SiO_(2)在示踪脂肪干细胞(adiposederived stem cell,ADSC)修复损伤面神经中的应用。方法制备并表征BaGdF_(5)@SiO_(2),用ADSC和UCNP共培养后得到ADSC-UCNP,观察UCNP在ADSC中的分布,通过CCK8细胞增殖试验、Transwell细胞迁移实验研究UCNP对ADSC增殖和迁移的影响,通过ADSC成骨诱导分化、成脂诱导分化探究材料对ADSC多向分化的影响。大鼠面神经损伤后尾静脉注射ADSC-UCNP,第1、7天时使用MicroCT对大鼠体内ADSC-UCNP的迁移、归巢进行实时跟踪。结果合适浓度的BaGdF_(5)@SiO_(2)对ADSC的增殖、迁移无明显影响,对ADSC成骨分化、成脂分化无明显影响,通过MicroCT在第7天成功观察到迁移到面神经损伤处的ADSC-UCNP,实现ADSC在大鼠体内修复面神经的实时影像学观察。结论 BaGdF_(5)@SiO_(2)具有良好的生物相容性,可用于ADSC在体内修复损伤面神经的实时示踪。Objective To explore the application of upconversion nanoparticles(UCNPs)BaGdF_(5)@SiO_(2)in tracking adipose derived stem cells(ADSCs)to repair facial nerve.Methods BaGdF_(5)@SiO_(2)was synthesized and characterized.ADSC-UCNP complex was obtained after ADSCs and UCNPs cocultivation.The distribution of UCNPs in the ADSCs was observed through electron microscope.The influence of UCNPs on ADSC proliferation and migration was explored through CCK8 assay,cell apoptosis assays and Transwell migration assay.The effects of UCNPs on ADSC multi-directional differentiation was explored through ADSC osteogenetic differentiation and adipogenic differentiation.The migration and homing of ADSC-UCNPs were tracked through MicroCT scanning on day 1 and 7 after injecting ADSC-UCNPs in facial nerve injury rats.Results The proper concentration of BaGdF_(5)@SiO_(2)had no obvious effect on the ADSCs proliferation and migration,and there was no effect on osteogenic differentiation and adipogenic differentiation.ADSC-UCNPs migrating to theinjury facial nerve site was successfully observed through MicroCT on the 7th day,and the real-time imaging observation of ADSCs in rats was realized.Conclusion BaGdF_(5)@SiO_(2)has good biocompatibility and can be used for tracking ADSC to repair facial nerve in vivo.
