慢病毒载体介导的circ_0091581通过靶向miR-1256调控肝癌细胞增殖、凋亡和侵袭迁移的机制

摘　　要：目的探讨circ_0091581对肝癌细胞增殖、凋亡和侵袭迁移的影响及分子机制。方法选取武汉市第四医院20例肝癌患者的癌组织及癌旁组织,用实时荧光定量PCR(RT-qPCR)检测circ_0091581和miR-1256的表达水平。Huh7细胞分为si-NC组、si-circ_0091581组、miR-NC组、miR-1256组、si-circ_0091581+anti-miR-NC组、si-circ_0091581+anti-miR-1256组,四甲基偶氮唑盐比色法(MTT)检测细胞活性;Transwell检测细胞迁移和侵袭;流式细胞术检测细胞凋亡;双荧光素酶报告实验验证circ_0091581和miR-1256的靶向关系。结果肝癌组织中circ_0091581表达水平升高,miR-1256表达水平降低(均P<0.05)。抑制circ_0091581表达或过表达miR-1256,Huh7细胞活性降低,迁移侵袭细胞数减少,细胞凋亡率升高(均P<0.05)。circ_0091581靶向调控miR-1256,下调miR-1256表达逆转了抑制circ_0091581表达对肝癌Huh7细胞增殖、迁移侵袭和凋亡的作用。结论抑制circ_0091581表达可能通过上调miR-1256抑制肝癌细胞增殖、侵袭迁移,促进其凋亡。Objective To explore the effect of circ0091581 on the proliferation,apoptosis,invasion and migration of liver cancer cells and its molecular mechanism.Methods The cancer tissues and adjacent tissues of 20 liver cancer patients in Wuhan Fourth Hospital were selected,and the expression levels of circ0091581 and miR-1256 were detected by real-time fluorescent quantitative PCR(RT-qPCR).Huh7 cells were divided into si-NC group,si-circ0091581 group,miR-NC group,miR-1256 group,si-circ0091581+anti-miR-NC group,si-circ0091581+anti-miR-1256 group.Tetramethylazolium salt colorimetric method(MTT)was used to detect cell activity;Transwell was used to detect cell numbers of migration and invasion;flow cytometry was used to detect cell apoptosis;dual luciferase report experiment was used to verify the targeting relationship between circ0091581 and miR-1256.Results The expression of circ0091581 in liver cancer tissue was increased,and the expression of miR-1256 was decreased(all P<0.05).Inhibition of circ0091581 expression or over-expression of miR-1256 significantly decreased Huh7 cell activity and the numbers of migrating and invaded cells,but increased apoptosis rate(all P<0.05).circ0091581 targeted and regulated miR-1256,down-regulation of miR-1256 expression reversed the effect of inhibiting the expression of circ0091581 on the proliferation,migration,invasion and apoptosis of liver cancer Huh7 cells.Conclusion Inhibiting the expression of circ0091581 may inhibit the proliferation,invasion,migration of liver cancer cells and promote apoptosis by up-regulating miR-1256.
