摘 要:目的通过相关分子生物学手段了解重要的内质网蛋白之一——内质网蛋白29(endoplasmic reticulum protein 29,ERp29)对人上皮性卵巢癌细胞顺铂化疗敏感性的影响。方法通过免疫组织化学法检测人卵巢癌组织中ERp29的表达情况。利用分子生物学手段下调ERp29的表达,RT-PCR及Western blot验证其表达水平,CCK-8及流式细胞术(FACS)分析上述卵巢癌细胞株顺铂敏感性及细胞周期等改变。结果在化疗耐药和化疗敏感的上皮性卵巢癌组织中,ERp29的表达水平存在较为显著的差异(P<0.05);转染ERp29-siRNA后,ERp29 mRNA和蛋白表达水平均显著降低,与对照组相比,差异均具有统计学意义(均P<0.05);CCK-8及FACS检测结果提示:转染ERp29-siRNA后,A2780细胞对顺铂的敏感性明显下降(P<0.05);FACS检测结果显示:顺铂(4μmol/L)处理细胞24 h后,与对照组相比,ERp29-siRNA组G_0/G_1期细胞比例明显降低,与对照组相比差异有统计学意义(P<0.05);S期及G_2/M期细胞比例增加,与对照组相比差异均具有统计学意义(均P<0.05)。结论ERp29可能在人上皮性卵巢癌顺铂化疗耐药过程中发挥重要作用。Objective Previous studies have found that ER stress may be involved in chemotherapeutic resistance as a protective mechanism of tumor cells.Our research attempts to explore the influence of endoplasmic reticulum 29(ERp29),one of the important endoplasmic reticulum proteins,on cisplatin sensitivity of human ovarian cancer cells through relevant molecular biological methods.Methods The expression of ERp29 in ovarian cancer tissue was detected by immunohistochemistry,the expression level of ERp29 in human ovarian epithelial cell line A2780 was down-regulated by molecular biological means,and its expression level was verified by RT-PCR and Western blotting.The cisplatin sensitivity and cell cycle changes of the above ovarian cancer cell lines were analyzed by CCK-8 and Fluorescence activated Cell Sorting(FACS).Results Expression of ERp29 was significantly different between chemotherapy drug resistant and chemotherapy drug sensitive ovarian epithelial cells.After transfection with ERp29-siRNA,the expression levels of ERp29 mRNA and protein decreased significantly as compared with the control group(both P<0.05).The results of CCK-8 and FACS indicated that after transfection with ERp29-siRNA,the sensitivity of A2780 cells to cisplatin decreased significantly,with statistically significant difference(P<0.05).The FACS showed that after treatment with cisplatin(4μmol/L)for 24 h,the cell proportion at stage G0/G1 in ERp29-siRNA group decreased,and both the proportion in stage G2/M and S increased,with statistically significant differences(both P<0.05).Conclusion ERp29 may play an important role in cisplatin resistance in epithelial ovarian cancer.
